半夏泻心方配伍药组含药血清对人胃癌细胞凋亡及凋亡相关基因的影响

摘　　要：目的研究半夏泻心方不同配伍药组含药血清对人胃癌BGC-823细胞凋亡及凋亡相关基因的影响。方法采用S-P免疫组化法检测凋亡相关基因bcl-2、bax的表达。结果半夏泻心方配伍中辛开组、苦降组和由其组成的辛苦组凋亡指数显著高于辛甘组、苦甘组、甘补组和全方组(P<0.05),呈剂量-效应关系。辛苦组可显著下调bcl-2基因的表达,上调bax基因的表达。结论半夏泻心方中辛开、苦降的配伍形式可能通过促进bax的蛋白表达、抑制bcl-2蛋白表达达到诱导细胞凋亡作用。辛开、苦降配伍后(辛苦)有显著的协同增效趋势,而其它配伍药群可能存在拮抗作用。提示辛开苦降法可能是中医药防治胃癌的主要治则治法之一。Objective To study effect of the components in serum of Banxia Xiexin Prescription and different herbal formula on the apoptosis of cell BGC-823 and apoptosis-related gene expression. Methods The genic expression of bcl-2 and bax was tested by S-P immunohistochemistry technique. The apoptotic index of the groups of Xinku in Banxia Xiexin Prescription was significantly higer than that in the groups of Xingan, Kugan, Ganbu and the intact prescription （P〈0.05）, respectively, it was related with the does and time. Moreover, the expression of bcl-2 down-regulated and bax up-regulated in the groups of Xinku. Conclusion The groups of Xinkai and Kujiang in Banxia Xiexin Prescription could induce the apoptosis of cell BGC-823, one of the mechanisms may be down-regulating of the bcl-2 and up-regulating of the bax genes expression. It was positive and cooperative during the compatibility of Xinkai and Kujiang. However, it was antagonistic during others compatibility. It suggested that the way of Xinkai and Kujiang was important for cure and prevention of gastric cancer.
