摘  要:用硫酸铵盐析法从抗猪瘟高免血清中提取免疫球蛋白,应用搅拌标记异硫氰酸荧光素(FITC),葡聚糖凝胶G-25层析除去标记物中游离的荧光素,制备出猪瘟荧光抗体(CSF-FA)。经测定,标记荧光抗体的染色效价为1∶4。用此浓度荧光抗体,结合常规诊断检测猪瘟,从33例采自陕西省不同地区自然发病猪的扁桃体中检出猪瘟阳性病例7份,阳性率21.3%(7/33)。结果表明,制备的荧光抗体诊断猪瘟敏感性高、特异性强;陕西省猪瘟自然感染发病率高。Immunoglobulin was extracted from swine serum against CSF using (NH4)2SO4 fractionation and labelled with fluorescein isothiocyanate (FITC) using agitation methods. Dissociative FITC was removed by Sephadex G-25 column. The working dilution of CSF-FA was 1 : 4. A total of 33 field cases from different places in Shaanxi province were tested for CSFV using direct fluorescent antibody. 7 positive cases were found from 33 field cases. The positive rate amounts to 21.3% (7/33). The technique possess highly specificity and sensitivity. The result indicated that CSFV infection would be prevalent in the pig herds of Shaanxi province.
