摘  要:以人脐静脉内皮细胞(Human umbilical vein endothelial cell, HUVEC)培养的方法,探讨了冠心舒和低分子右旋糖酐抗氧自由基和多聚阳离子致HUVEC损伤的作用。结果表明,冠心舒可对抗氧自由基和多聚阳离子对HUVEC的损伤;低分子右旋糖酐只能对抗氧自由基对HUVEC的损伤,而不能对抗多聚阳离子的损伤作用。In this article, the protective effects of Cuaimnshu and low molecular weight dextran (dextran 40) on HUVEC injury were studied. In the first experiment, cells were damaged by the addition of xanthine and xanthine oxidase. As a parameter of cell injury, the activity of LDH was examined. The results showed that Guanxinsfm and dextran 40 significantly decreased the release of LDH from cells (P<0. 05-0. 01) , and the action of Guanxinshu was stronger than that of dextran 40 (P<0. 05-0. 01). In the second experiment, cells were damaged by adding polylysine (MW 50 000) to media and cell injury was assessed by 3H-TdR incorporation. We found that Guanxwshu could significantly antagonize polycation-mediated cell damage (P<0. 01) , but dextran 40 showed no effect.These results suggested that glycosaminoglycans could protect HUVEC from injury of oxygen-free radicals and polycations, and that the protective effects seemed to depend on the degree of sulfation.The inhibition effect of GtianxuisJu on cell growth, as shown in experiment 3, implied that the protective actions were not due to cell proliferation. The mechanisms of these actions are still unclear.
