摘 要:目的观察H2S对血管一氧化氮(NO)释放的影响,以探讨H2S和NO这两种气体信号分子间的相互作用。方法大鼠腹腔注射NaHS(14μmol/kg/d,分3次),3d后取血液测定血浆亚硝酸盐含量。离体孵育大鼠主动脉薄片,加入NaHS(10-7 ̄10-4mol/L)孵育4h;及50μmol/LNaHS分别孵育2、4、6h。采用Greiss法测定血管孵育液亚硝酸盐含量。结果NaHS腹腔注射组大鼠血浆NO2-含量较对照组减少21%(P<0.01)。大鼠主动脉薄片离体孵育,NaHS(10-7 ̄-10-4mol/L),呈浓度依赖的抑制了血管一氧化氮释放,NO2-含量分别较对照组低10%(P<0.05)、30%、38%和42%(P<0.01),IC50为0.499μmol/L;而一次性给予50μmol/LNaHS,孵育2h、4h和6h,孵育液中NO2-含量比对照组降低62%、40%(P<0.01)和19%(P<0.05)。结论H2S减少血管NO生成。[Objective] To observe the influence of nitric oxide induced by hydrogen sulfide (H2S), to explore the interaction of H2S and NO as cardiovascular regulatory gasotransmitters. [Methods] Eight Male Sprague -Dawley rats were divided into 2 groups (Naris peritoneal injection groups and normal groups, n=4/group). The content of NO2^- in plasma were determined. Administrated NariS (10^-7 mol/L~10^-4 mol/L), a donor of H2S, incubation for 4 hours, or 50 μmol/L Naris incubation for 2 h, 4 h and 6 h, respectively, in aortic thin slices in vitro. Measured the nitrite production of aorta by Greiss assay. [Results] After 3 days injection of NariS, the plasma level of NO2^- of rats in Naris group decreased by 21% compared to control group (4.71±0.47) umol/L. After giving Naris (50 μmol/L) one time, and incubating for 2 h, 4 h, the nitrite production decreased by 62%, 40% (P 〈0.01); and after incubation for 6 h, the nitrite production still was inhibited by 19% (P 〈0.05). NariS (10^-7 mol/L~10^-4 mol/L) inhibited the nitrite production by 10%(P 〈0.05), 30%, 38% and 42% (P 〈0.01) as a dose-dependent manner, and IC50 were 0 409 μmol/L. [Conclusion] H2S can inhibit NO release of aortic tissue.
