摘 要:目的探讨单侧输尿管梗阻大鼠肾小管间质P38MAPK、磷酸化P38MAPK的表达变化,从而阐明P38MAPK在肾小管间质纤维化发生中的作用。方法将30只雌性SD大鼠随机分为假手术组及模型组,模型组行UUO术。术后第5、10、15天分别处死两组中的5只大鼠,取左肾行HE和Masson染色,并用免疫组化方法检测肾小管间质中TGF-β1、P38MAPK、P-P38MAPK的表达。结果模型组第5天组肾小管间质增宽,小管间质细胞增多,梗阻后第10天病变明显,第15天可见弥漫的肾小管基底膜增厚皱缩,纤维化明显(P<0.01)。免疫组化结果显示:假手术组大鼠肾小管间质中仅有少量TGF-β1、P38MAPK和P-P38MAPK表达。模型组大鼠梗阻侧肾小管间质中TGF-β1、P38MAPK和P-P38MAPK表达明显增加(P<0.01),随时间进展,表达更加明显。结论实验大鼠梗阻侧肾小管间质中TGF-β1、P38MAPK和P-P38MAPK表达上调,P38MAPK信号通路可能参与UUO大鼠肾小管间质纤维化过程。[Objective] To observe the expression of transforming growth factor-β1 (TGF-β1), P38MAPK and P- P38MAPK in renal tubular interstitium of UUO rats. [Methods] Thirty female Sprague Dawley rats were divided into two groups: the sham-operated group and the model group. The model group's left ureter was dissociated to be obstructed. The sham-operated group was dissociated left ureter only, and not to be obstructed. Five rats of each group were killed respectively at 5, 10, 15 d after surgery. Histological changes of renal tubular interstitium was observed by HE and Masson staining. Iminunohistochemistry was performed on left renal tubular interstitium for TGF- β1, P38MAPK and P-P38MAPK in renal tubular interstitium at each time point. [Results] The representative appearance of HE staining and Masson's triehrome-staining indicating interstitial expansion and fibrosis of obstructed kidneys was prominent in the model group at each time point of UUO rats as compared to control group (P 〈0.01). TGF-β1, P38MAPK and P-P38MAPK were faintly stained in the sham-operated kidneys. Immunostaining of TGF- β1, P38MAPK and P-P38MAPK in renal tubular interstitium increased progressively starting from 5 to 15 d after UUO. [Conclusion] The expression of P38MAPK in renal tubular interstitium of UUO rats was increased significandy, P38MAPK maybe take part in kidney fibrosis.
