摘 要:建立了HPLC测定红霉素血药浓度的新方法。以峰面积定量,浓度与峰面积具有良好相关性(r=0.998),线性范围0.4~4.0μg/ml,最低检测限0.1μg/ml。浓度为0.4,1.6,3.2μg/ml的回收率均大于93.4%,三个浓度下测定的日内差及日间差(RSD)分别为1.23%,1.49%,1.67%和2.18%,3.26%,4.17%,与微生物法测定结果无显著差异。A high performance liquid chromatographic method for the determination of erythromycine in plasma is described. The method comprises the extraction from the plasma samples, and the base catalyzed decyclisation. The reaction product was detected by a reversed phase HPLC system installed with UV measurements at 235.5±1 nm. The linear range of concentration was 0.4 to 4.0 μg/ml with the correlation coefficient 0.998. The minimal determinable concentration was 0.1 μg/ml. At the three concentrations of 0.4, 1.6, 3.2 μg/ml, the recoveries were above 93.4% ; the intra and inter day variation coefficient were 2.18%,3.26%,4.17% and 1.23% , 1.49% ,1.67% respectively. The result of the microbiologic method agreed with that of the HPLC method.
