摘 要:目的观察甲状腺功能亢进(甲亢)大鼠中枢神经系统Ⅱ型脱碘酶mRNA(D2-mRNA)的表达。方法将Wistar大鼠分为对照组和甲亢组,甲亢组采用40 g/L甲状腺素灌胃。连续3周后定量荧光RT-PCR技术分析甲亢大鼠大脑、小脑、海马、脑干和脊髓中D2-mRNA表达的变化。结果甲亢大鼠大脑、小脑、海马、脑干和脊髓的D2-mRNA的表达均被抑制,与对照组比较,分别下调22.21%、45.57%、31.56%、34.79%、85.10%。结论甲亢时大鼠中枢神经系统的D2在转录水平受到抑制,说明D2对中枢神经系统自稳态的调节起着重要的作用。Objective To detect the expression of type 2 iodothyronine deiodinase mRNA in hyperthyroid central nervous system. Methods Wister rats were divided into normal group and hyperthyroid group. The expression of type 2 deiodinase in cerebrum, cerebellum, hippocampus, brain stem and spinal cord of hyperthyroid rats was measured by real-time PCR after 3 weeks intragastric administration of 40 g/L tetraiodothyronine for hyperthyroid group. Results At transcriptional level,the expression of type 2 iodothyronine deiodinase was obviously inhibited in cerebrum, cerebellum, hippocampus, brain stem and spinal cord of hyperthyroid rats compared with those of normal control group, decreasing by 22.21%, 45.57%, 31.56%, 34.79% and 85.10%, respectively. Conclusions The type 2 iodothyronine deiodinase in the central nervous system of hyperthyroid rats is inhibited at transcriptional level, indicating that type 2 iodothyronine deiodinase plays an important role in adjusting the homeostasis of central nervous system.
