摘 要:模拟天然皮肤的真、表皮结构,以胶原和壳聚糖为主要原料构建皮肤组织工程双层支架.将固含量0.8%(mass)的胶原-壳聚糖溶液冻干后压膜,经戊二醛交联后再冻干形成双层支架的底层,然后注入固含量0.2%(mass)的胶原-壳聚糖溶液,经冻干、交联、再冻干即得结构较稳定的双层复合海绵支架.在双层支架上培养的小鼠成纤维细胞贴壁生长正常.本研究构建的胶原-壳聚糖双层支架具有良好的结构稳定性和细胞相容性.To simulate the natural skin composed of dermis and epidermis,a bilayer collagen-chitosan scaffold was developed. The bilayer collagen-chitosan scaffold was prepared by lyophilization of 0.8% (mass) collagen-chitosan solution, then it was pressed by 15 kg weight for 24 h,and cross-linked by 0.2% (mass) glutaraldehyde at 4℃for 24 h. After the scaffold was freeze-dried,0.2%(mass) collagen-chitosan solution was infused,freeze-dried,cross-linked by 0.2% (mass) glutaraldehyde at 422 for 24 h, freeze-dried again. The microstructure and cytocompatibility of the scaffolds were investigated. The contraction ratio enhanced with the increase of content of collagen-chitosan solution. The contraction ratio was reduced after 24 h of pressure. The surface layer of the scaffold prepared by 0.8 % (mass) collagen-ehitosan solution improved the adhesion of cells. The inner layer of the scaffold prepared by 0.2 % (mass) collagen-chitosan solution promoted the proliferation of cells. The bilayer collagen-chitosan scaffold has good cytocompatibility and structure stability. Thus, it is suitable as scaffold for skin tissue engineering.
