摘　　要：对虾白斑综合症病毒是一种新型的DNA病毒.VP24蛋白质是该病毒粒子的一个主要的结构蛋白质.当纯化的病毒粒子经去污剂处理后,VP24蛋白质被发现完全存在于囊膜部分,Western杂交实验也证实该结果,以上实验结果表明VP24蛋白质是病毒的一个囊膜蛋白质.利用免疫胶体金定位技术对该蛋白质的进一步研究中发现,核衣壳上未见标记的金颗粒,完整的病毒粒子上标记的金颗粒也很少,而囊膜部分破损的病毒粒子上却可观察到大量的金颗粒.我们认为VP24蛋白质是一个囊膜蛋白质,但是它的主体可能存在于病毒的囊膜和核衣壳之间.VP24蛋白质的定位研究将加速阐明病毒感染和包装的机制,并将有助于病毒的诊断和控制.White Spot Syndrome Virus （WSSV） represents a new genus of DNA viruses. We described herein the characterization of VP24,one of major structural proteins of WSSV. When purified virions were treated with Triton X-100 detergent, VP24 was found to present exclusively in the envelope fraction, which was also confirmed by Western blot analysis. The result indicated that VP24 behaved as an envelope protein. Immunoelectron microscopy showed that very small amounts of labelling were observed on intact virions,whereas a small proportion of particles which were disrupted by the preparation were labelled strongly. We have rationalized this finding by suggesting that the main body of VP24 might be present in the space between the envelope and the nucleocapsid. The exploration of VP24 localization will facilitate a better understanding of the molecular mechanism underlying WSSV infection and assembly,and may be helpful for the diagnosis and control of virus infection.
