蛴螬粗提物对人宫颈癌Hela细胞抑制作用的形态学观察

摘　　要：目的研究蛴螬粗提物对体外培养的人宫颈癌Hela细胞株抑制作用引起的形态学改变。方法应用MTT法、HE染色、油红O染色、透射电镜的方法检测蛴螬粗提取物对体外培养的人宫颈癌Hela细胞抑制作用的影响及观察细胞的形态学改变。结果蛴螬3种粗提取物中以石油醚粗提物增殖抑制率最高。蛴螬石油醚粗提物对人宫颈癌Hela细胞株的增殖抑制作用呈剂量依赖性(P<0.01),同时呈时间依赖性(P<0.01)。倒置显微镜下蛴螬石油醚粗提物50μg/m l作用后早期细胞出现凋亡形态学变化;作用晚期细胞发生膜破裂坏死。HE染色和透射电镜观察细胞出现典型凋亡形态变化。油红O染色和透射电镜观察到细胞发生脂肪变性。结论蛴螬粗提物对人宫颈癌Hela细胞株具有抑制增殖及诱导凋亡作用。蛴螬3种粗提物中以石油醚粗提物抑制增殖作用效果最佳。蛴螬石油醚粗提物对Hela细胞抑制增殖作用在粗提物50μg/m l作用48 h之前主要以诱导凋亡为主,在其后时间以诱发肿瘤细胞脂肪变性和坏死为主。Objective To observe morphological changes of the effect of the apoptosis and degeneration of Hela human cervical carcinoma cells in vitro after treatment with the raw extract of Grub, Methods The growth inhibition rates and cytotoxic effects of Hela cells were measured with MTT assay. Observing cells under inverted microscope, further testing the changed morphology by HE and ＂O＂ red oil stains optical microscope. Observing the uhrastructure changes of Hela with transmission electron microscope, Results The Grub petroleum ether extract could significantly inhibit the growth and proliferation of Hela cells than the other extracts, and the effects were in a dose - dependent and time - dependent manner, Under inverted microscope the Hela cells showed the apoptosis changes by being treated with 50 μg/ml of the Grub petroleum ether extract before 48 hours ; after 48 hours cells showed necrosis. The cells showed classic morphological features of apoptosis and fatty degeneration under light microscope and electronic microscope. Conclusion The raw Grub extract has inhibition of proliferation and inducing apoptosis for Hela cells in vitro; and the effect of petroleum ether extract was the best. Before being treated with the 50 μg/ml of the Grub petroleum ether extract for 48 hours, the effect of inhibitive proliferation mainly was completed by the induction of apoptosis ; after 48 hours the effect was completed by the induction of fatty degeneration and necrosis.
