摘  要:目的:建立贞芪扶正胶囊(女贞子,黄芪等)中黄芪甲苷和红景天苷的含量测定方法。方法:Inertsil ODS-3 C18(250mm×4.6 mm,5μm)色谱柱,黄芪甲苷采用HPLC-ELSD法,流动相为乙腈-水(37∶63),流速1.0 mL/m in,ELSD检测器漂移管温度106°C,载气(空气)流速2.7 L/m in。红景天苷采用反相高效液相色谱法,以乙腈-水为流动相低压梯度洗脱,流速1.0 mL/m in;检测波长278 nm。结果:在色谱试验条件下,黄芪甲苷在0.76μg^3.81μg范围内呈良好的线性,R=0.999 5;平均加样回收率为98.4%,RSD=0.9%(n=6)。红景天苷在0.4~4μg范围内呈良好的线性,R=0.999 9;平均加样回收率为100.7%,RSD=2.1%(n=5)。结论:方法简单,准确,灵敏,能有效控制贞芪扶正胶囊的质量。AIM: To establish the method of determining astragaloside Ⅳ and salidroside in Zhenqi Fuzheng Capsule (Fructus Ligustri Lucidi, Radix Astragali, etc. ). METHODS: Astragaloside Ⅳ was determined by HPLC-ELSD on Inertsil ODS-3 C18 column (250 mm×4.6 mm, 5μm) using a mobile phase of acetonitrile-water (37: 63). The flow rate was 1.0 mL/min and detector parameters were set as follows: drift tube temprature was at 106℃ ; carrier gas (air) flow rate was 2.7 L/min. Salidroside was determined by RP-HPLC on the same column using a mobile phase composed of acetonitrile-water with gradient elution. The detection wavelength was at 278 nm and the flow rate was 1.0 mL/min. RESULTS : Calibration curve of astragaloside Ⅳ was linear between 0.76μg and 3.8μg under the chromatographic condition, R = 0. 9995. The average recovery of astragaloside Ⅳ was 98.4%. RSD was 0.9% ( n = 6). And the calibration curve of Salidroside was linear between 0.4μg and 4μg, R =0.999 9. The average recovery of salidroside was 100.7%. RSD was 2.1% (n =5). CONCLUSION: These methods were simple, accurate and sensitive, so it can be used for the quality control of Zhenqi Fuzheng Capsule.
