摘 要:目的研究内蒙古地区汉族人群IgA肾病与TNFβ基因多态性的关系。方法用聚合酶链反应-限制性片段长度多态性(PCR-RFLP)技术对79例IgA肾病患者及80例正常对照TNFβ1 069位点等位基因A→G单碱基突变多态性进行分析。结果内蒙古地区汉族IgA肾病患者TNFβ*1/1纯合子和TNFβ*2/2纯合子检出率与正常对照组比较均有较大差异(均P< 0.05);TNFβ*2等位基因频率(68.99%)较正常人(53.75%)显著升高(P<0.01)。结论TNFβ*2基因可能与IgA肾病易患性相关。Objective To study the relationship of tumor necrosis factor beta (TNF-β) polymorphism with human IgA nephropathy ofChinese Han nationality in Inner Mongolia Autonomous Region. Methods The A→G single base mutation polymorphism in TNF-β1096 locus were analyzed among 80 normal controls and 79 IgA nephropathy patients by using polymerase chain reaction-restriction fragmentlength polymorphism (PCR-RFLP). Results The genotype frequencies of TNFβ * 1/1 and TNFβ * 2/2 in IgA nephropathy patientswere significantly higher than that in normal controls (X^1/1 = 5.58 ,P 〈 0.01;X^2 2/2 =4.76,P 〈0.01 ). The gene frequency of the TNFβ 2 in IgA nephropathy patients (68.99%) was significantly higher than that in normal controls (53.75 %, X2 = 7.78, P 〈 0.01,R^1 =1.91 ). Condusion TNFβ* 2 allelic frequency is associated with IgA nephropathy.
