痤疮丙酸杆菌培养滤液对THP-1细胞增殖和吞噬功能的影响

摘　　要：目的:探讨外伤后细菌性致死性肉芽肿病原菌-痤疮丙酸杆菌培养滤液对单核巨噬细胞株THP-1细胞增殖、吞噬功能的影响。方法:采用MTT比色法、DAF比色法观察比较痤疮丙酸杆菌标准株与从本病患者皮损分离菌株在相同浓度培养滤液及相同菌株不同浓度的培养滤液对THP-1细胞增殖、吞噬功能的影响。结果:1、痤疮丙酸杆菌培养滤液作用于THP-1细胞,可见细胞在12h时增殖明显,48h增殖达到高峰,和空白对照组相比有显著性差异(P<0.05);而标准株较患者分离株培养滤液的刺激增殖作用强(P<0.05);2、培养滤液均能促进THP-1细胞的吞噬功能,并且促进作用的强弱与滤液浓度有关。标准株在20%浓度时刺激作用最强,分离株均表现为100%浓度时刺激作用最强。但各菌株滤液间无显著性差异(P>0.05);结论:本病致病菌及标准株培养滤液对THP-1细胞均有明显促进增殖的作用;对THP-1细胞有促吞噬作用且与浓度有关。Objective To research the change of multiplication, phagocytosis of THP-1 cells by Propionibacterium acnes （P.acnes）,which is the pathogen of Fatal Bacterial Granuloma after Trauma. Methods MTT colorimetry, DAF colorimetry were used to research regulatory role of THP-1 cells by filtrate of P.ances cultureon THP-1 cells after the treatment of filtrate of P.ances culture. Results Filtrate of P.ances culture significantly enhanced the multiplication of THP-1 cells （P〈0.05）. Different strains P.ance have different capabilities,the capability of control stain is more powerful than other stains which divided from patients. And the control stain had more significant effect on THP-1 cells than other stains dividing from patients （P〈0.05）. And filtrate of P.ances culture also enhanced the phagocytosis of THP-1 cells. It depended on the concentration of filtrate of P.ances culture. Conclusion： Different P.ances stains have different capacities of enhancing multiplication and phagocytosis of THP-1 cells.
