用高效液相色谱法测定褪黑激素

摘　　要：本文首次用高效液相色谱（紫外检测器）测定褪黑激素（ｍｅｌａｔｏｎｉｎ）。检测波长２６０ｎｍ，流动相为甲醇∶Ｈ２Ｏ＝１∶１（Ｖ／Ｖ），（ｐＨ４．０），流速１ｍｌ／ｍｉｎ。研究结果，样品重复性（峰面积）为±δｘ＝４８７３２．２０±２５７．６８，ＣＶ％为０．５３％。结果表明，利用高效液相色谱（ＨＰＬＣ）测定褪黑激素远比目前应用的放射免疫试验、免疫组织化学法和气谱－质谱法简便、快速。High performance liquid chromatography (HPLC) of melatonin (standard and our synthetic) was carried out on Lichrosorb RP-18 column (controlled at 30℃),the compounds were determined by HPLC (ultraviolet detector) .Detective wavelength was 260nm.The mobile phase was mathanol:water=11 (pH4.0) ,the flow rate was 1ml/min. The study showed that repetition of melatonin was satisfactory ( CV%0.53%～0.87%) .rate of recovery was 100.53%～106.60%.The study also showed that this method was far more buperior than most of thoes used at present.
