摘 要:目的:观察神经降压素(NT)对实验性神经细胞老化过程中细胞周期,DNA和细胞总蛋白的影响。方法:采用小鼠神经母细胞瘤细胞无血清培养建立神经细胞老化实验研究模型。以流式细胞光度术测定的细胞周期,DNA和细胞总蛋白为实验性神经细胞老化的指标,观察NT对这些指标的影响。结果:NT促使滞留于G1期的老化细胞向S期转化并降低伴随培养时间延长而增高的细胞总蛋白。结论:NT是通过使细胞周期、DNA和细胞总蛋白发生变化来发挥其延缓实验性神经细胞老化的作用。To observe the effects of neurotensin on cell cycle and total cellular protein during experimental neuronal aging.METHODS:Serum-free culture of mouse neuroblastoma cells was used as the experimental model for the study of neuronal aging,with flow cytometry of cell cycle,DNA and total cellular protein as the indices of neuronal aging.After addition of NT 10-7 mol/L into the culture medium,the influences of NT on the indices were studied.RESULTS:(1)NT could increase the number of S and G_2+ M phase cells,and decrease the number of G_1 phase cells.(2)NT could decrease total cellular protein in aged experimental neural cells. CONCLUSION:NT produces effects on cell cycle,DNA and total cellular protein in the direction of delaying neuronal aging.The interrelationship between NT and neuronal aging merits further investigation.
