Determination of Alkaloids in Lycoris radiata with Microwave-assisted Extraction Coupled with High Performance Liquid Chromatography

摘  要:采用微波辅助提取法提取石蒜中的石蒜碱、力可拉敏和加兰他敏生物碱,建立了微波辅助提取/HPLC测定石蒜中这3种生物碱的分析方法。用正交试验设计结合单因素试验优化了样品颗粒度、提取温度、提取时间、固液比等微波提取条件。采用三乙胺水溶液(pH9.0)-乙腈-甲醇作为流动相对石蒜碱、力可拉敏和加兰他敏进行HPLC测定,方法的线性范围为5.0—400mg/L,检出限分别为5.0、5.1、10.2ng,RSD分别为2.2%、5.7%和4.4%。与传统的溶剂回流法比较,MAE提取法快速、高效。用本法分析了10个不同产地石蒜中石蒜碱、力可拉敏和加兰他敏。Lycorine, lycoramine and galanthamine in Lycoris radiata were microwave-assisted extracted (MAE) and determined by HPLC using triethylamine aqueous solution ( pH 9.00 ) - acetonitrile - methanol as the mobile phase. The MAE operating conditions were optimized by the orthogonal test design combined with the mono-factor test. The linear ranges of the calibration curves for lycorine, lycoramine and galanthamine were 5.0 -400 mg/L, respectively. Their detection limits were 5.0, 5.1 and 10.2 ng, respectively and the RSD were 2.2% , 5.7% and 4.4% , respectively. Compared with the results obtained by the conventional solvent reflux extraction method, the MAE was time saving and more efficient. The method was successfully applied to the extraction and determination of Lycorine, lycoramine and galanthamine in 10 Lycoris radiata samples from different regions.
