摘  要:目的探讨血红素氧合酶-1(HO-1)高表达抗兔心肌IR损伤的效果及其作用机制。方法32只兔随机均分为单纯IR组与HO-1诱导组,分别预先给予生理盐水5mL及血晶素25mg/kg腹腔内注入,24h后,开胸结扎冠脉左前降支缺血40min,松开再灌注4h,测定心肌中HO-1表达及活性、中性粒细胞浸润、丙二醛(MDA)含量、心肌梗死范围及心肌细胞凋亡指数。结果与单纯IR组比较,HO-1预诱导组心肌中HO-1表达及活性均明显升高(P<0.01),中性粒细胞浸润及MDA含量明显降低(P<0.01),心肌梗死范围及心肌细胞凋亡指数均显著减少(P<0.01)。结论HO-1可通过抗炎、抗氧化等作用减轻急性心肌IR损伤。[Objective] To explore the effects and mechanism of heme oxygenase-1 (HO-1) overexpression protection of rabbit heart from ischemia reprfusion(IR) injury. [Methods ] 32 rabbits were randomly divided into simple IR group and HO-1 induced group, each rabbit was intra-peritoneal injections of 5 mL saline in simple IR group and 25mg/kg heroin in HO-1 induced group respectively. 24 hours later, rabbit heart underwent ischemia for 40 minutes by ligatin left anterior descending coronary artery and reperfusion for 4 hours. The myocardial HO-1 expression and HO-1 activity, polymorphonuclear neutrophils(PMN) infiltration, MDA content, infarct size and the apoptosis index(AI) in both groups were detected. [Results] The amount and activity of myocardial HO-1 of HO-1 induced group were significantly higher than that of simple IR group (P 〈0.01 respectively). While the myocardial PMN infiltration, MDA content, infarct size and the AI of HO-1 induced group were much lower than that of simple IR group (P 〈0.01, respectively). [Conclusion] HO-1 can reduce myocardial IR injury by means of anti-oxidative and anti-inflammatory properities.
