镇脑宁胶囊治疗血管神经性头痛等功效与其所含金属元素的关系

摘　　要：目的:从中药中微量元素与中药功效的关系出发,分析治疗血管神经性头痛的中成药制剂镇脑宁胶囊的功效与其含有的金属元素的关系。方法:实验于1999-03/04在泰山医学院化学实验室、山东农业大学中心实验室完成。镇脑宁胶囊购自泰安市医药采购供应站熏由中国通化东宝药业股份有限公司生产(批号980120)。主要成分:川芎、藁本、细辛、白芷等。功能与主治:熄风通络。准确称取有代表性的粉末状中药3份(各约2g),消解后得待测液,用原子吸收分光光度法和用电感耦合等离子体发射光谱法测定待测液中镁、锌、铜等12种金属元素的含量,同时做空白实验和回收率实验。回收率(%)=(加入纯品后的测得量-加入前的测得量)/纯品的加入量×100%。利用测定结果、脑血管病的病理生理和临床资料,分析镇脑宁胶囊的功效和其含有的金属元素的关系。结果:①镇脑宁胶囊中铜、锌、锰、铁、钴、镍、铬7种元素的回收率为(95.6±4.5)%～(103.0±5.7)%,符合分析化学实验回收率的常规要求。②镇脑宁胶囊中镁、锌、铜、铬、锰、铁、锂、钙、钾、镍、钴、钒的含量依次如下:(1763.133±120.439),(32.260±1.218),(7.930±0.040),(3.135±0.021),(25.023±0.550),(493.333±31.991),(0.556±0.026),(3215.100±77.969),(8643.967±178.972),(2.430±0.151),(0.399±0.208,(0.855±0.069)μg/g。③铁、铬、锂、镁、铜、锌、锰的含量分别是陆生被子植物中铁、铬、锂、镁、铜、锌、锰含量的3.52,13.65,5.56,0.55,0.57,0.20,0.04倍,钙、铁、锌、镁、锰、铜、镍、钴、钒的含量分别是植物类中药中相应元素含量的5.74,1.10,1.19,0.79,0.55,0.68,1.38,0.66,0.66倍。结论:镇脑宁胶囊治疗血管神经性头痛等功效可能与其可以补充患者体内适量的铁、铬、锂、镁、铜、锌、锰元素和川芎嗪、藁本挥发油等有关。AIM： To explore the correlation between the function of zhennao ning capsule in the treatment of angioneurotic headache and its metallic elements based on the relation between trace elements in Chinese herbs and their functions. METHODS： The experiment was conducted in the Chemical Laboratory of Taishan Medical College and Center Laboratory of Shandong Agricultural University between March and April 1999. Zhennao ning capsule, purchased from the Tai＇an Purchase and Supply Station, produced by the Tonghua Dongbao Medicines Co., Ltd. （No, 980120）, was composed of szechean lovage rhizome, Chinese louage, manchurian wildginger, dahurian angelica root and other Chinese herbs. Its functions included calming the endopathic wind and removing the obstruction. After weighed accurately of 3 parts of the representative powdery of the Chinese herbs （2 g）, the extract was prepared by digesting to determine its contents of 12 kinds of metallic elements including magnesium （Mg）, zinc （Zn）, cuprum （Cu） and so on with methods of atom ＇absorption spectrophotometry and inductance coupling plasma emission spectrum, meanwhile, the blank test and recovery test was accomplished. Rate of recovery （%）=（amount after adding with pure druginitial amount）/added amount ×100%. The relation between function of zhennao ning capsule and the metallic trace elements in it was studied by measuring results, pathophysiological processes and clinical study of angioneurotic disease. RESULTS： ①The rate of recovery of the seven elements including Cu, Zn, manganese （Mn）, ferrum （Fe）, cobalt （Co）, nickel （Ni） and chromium （Cr） were （95.6±4.5）%-（103.0±5.7）%,which conformed to the common requirement in analytical chemistry. ②The content （μg/g） of Mg, Zn, Cu, Cr, Mn, Fe, Li, Ca, K, Ni, Co and V were （176 3.133±120.439）, （32.260±1.218）, （7.930±0.040）, （3.135±0.021）, （25.023±0.550）, （493.333±31.991）, （0.556±0.026）, （321 5.100±77.969）, （864 3
