摘  要:目的:观察中药方剂化痰活血方对高脂血症大鼠的调脂作用及含药血清对牛主动脉内皮细胞的保护作用。方法:整体实验于2002-03/2004-02在湖北中医学院中医临床基础学科实验室完成。选取健康Wistar大鼠60只,随机数字表法分为6组,空白组、高脂血症模型组、力平脂组、化痰活血方高、中、低剂量组,每组10只。采用高脂乳剂饲养10d诱导大鼠高脂血症模型,然后加用化痰活血方提取物喂养20d,然后检测大鼠血中总胆固醇、三酰甘油、低密度脂蛋白胆固醇、高密度脂蛋白胆固醇及一氧化氮水平;在原代培养的牛主动脉内皮细胞中加入化痰活血方提取物喂养的动物血清干预,并用MTT法和WesternBlot杂交技术分别观察化痰活血方含药血清对牛主动脉内皮细胞细胞增殖及内皮型一氧化氮合酶表达的影响。结果:纳入动物60只,均进入结果分析。①化痰活血方高、中、低剂量组均能显著降低高脂血症大鼠血中总胆固醇、三酰甘油、低密度脂蛋白胆固醇水平,显著提高高密度脂蛋白胆固醇、高密度脂蛋白胆固醇亚组分、一氧化氮水平,与高脂血症模型组比较差异有显著性意义眼总胆固醇分别为(1.73±0.07),(1.74±0.07),穴1.42±0.07雪,穴2.62±0.09雪mmol/L鸦三酰甘油分别为穴1.03±0.08雪,穴0.95±0.06雪,穴1.19±0.12雪,穴4.38±0.34雪mmol/L鸦低密度脂蛋白胆固醇分别为穴0.30±0.05雪,穴0.25±0.04雪,穴0.15±0.03雪,穴0.46±0.04雪mmol/L;高密度脂蛋白胆固醇分别为(0.99±0.06),(1.06±0.05),(0.89±0.10),(0.69±0.04)mmol/L;高密度脂蛋白胆固醇亚组分分别为(0.38±0.07),(0.39±0.05),(0.43±0.06),(0.24±0.05)mmol/L;一氧化氮分别为(16.3±1.39),(14.3±1.02),(15.6±2.47),(15±1.6)μmol/L,P<0.05或P<0.01演。②50,100,150g/L化痰活血方含药血清明显促进牛主动脉内皮细胞细胞增殖和内皮型一氧化氮合酶蛋白表达。结论:化痰活血方对高脂血症大鼠不仅有明显的�AIM: To observe the effect of Huatan Huoxue Fang (HHF) on plasma lipids in experimental rats with hypcrlipidemia in and the protective effect of serum containing drug on bovine aortic endothelium. METHODS: The experiment was conducted in the Laboratory of Clinic Elemental Science of Traditional Chinese Medicine, Hubei College of Traditional Chinese Medicine from March 2002 to February 2004. Sixty healthy Wistar rats were randomly divided into six groups (10 for each group): blank group, hyperlipidemia model group, lipanthyl group, the high dosage HHF-troated group, middle dosage HHF-treated group and low dosage HHF-treated group. The Wistar rats were fed with lipidemia emulsion for ten days to induce hyperlipidemia model and then were exposed to HHF prescription for twenty days. Plasma levels of total cholesterol (TC), triglyceride (TG), low-density lipoprotein cholesterol (LDL-C), high-density lipoprotein cholesterol (HDL-C), and nitric oxide (NO) were detected. Furthermore, bovine aortic endothelial ceils (BAEC) were isolated and cultured, and were incubated with the serum of rat treated with extract of HHF to determine the effects of HHF on cell proliferation and endothelial nitric oxide synthase (eNOS) expression by MTT assay and Western blot hybridization technique. RESULTS: A total of 60 included animals were involved in the result analysis. ①The high dosage HHF, the middle dosage HHF and the low dosage HHF obviously reduced the levels of TC, TG and LDL-C, and significantly increased the levels of HDL-C, HDL2-C and NO, which showed obvious difference from the hyperlipidemia model group [TC (1.73 ±0.07), (1.74±0.07),(1.42±0.07),(2.62±0.09) mmol/L, respectively; TG (1.03±0.08),(0.95±0.06),(1.19±0.12),(4.38±0.34) mmol/L, respeetivley; LDL-C (0.30±0.05),(0.25±0.04),(0.15±0.03),(0.46±0.04) mmol/L, respectively; HDL-C (0.99±0.06), (1.06±0.05), (0.89±0.10), (0.69±0.04) mmol/L, respecti
