摘  要:目的制备转化生长因子β1(transforminggrowthfactor-β1,TGF-β1)壳聚糖不对称膜,研究其对小鼠成牙本质细胞(MDPC-23)生长和增殖的影响。方法用自然干燥法和冷冻干燥法,复合TGF-β1壳聚糖微球,制备TGF-β1壳聚糖不对称膜,利用扫描电镜进行形态学观察。TGF-β1壳聚糖不对称膜与小鼠成牙本质细胞共培养,2d后扫描电镜观察细胞在材料上的黏附、生长,并绘制7d内的细胞生长曲线。结果壳聚糖不对称膜为双层结构,具有致密膜和多孔层,孔径60-160μm,孔隙率为83·26%,与水的结合能力为15·6g水/g多孔膜。TGF-β1壳聚糖微球均匀分布于多孔层的孔隙中。MDPC-23在TGF-β1壳聚糖不对称膜的多孔层黏附生长,细胞的增殖能力显著提高,培养7d,细胞数量由(2·03±0·04)×104增至(18·31±0·60)×104。结论壳聚糖不对称膜具有理想的物理性能,复合TGF-β1壳聚糖微球后形成TGF-β1三维缓释系统,可促进细胞的增殖。Objective To develop asymmetric porous chitosan membranes in combination with TGF-β1-loaded chitosan microspheres (MS-TGFs) and evaluate the effect of released TGF-β1 on the proliferation potential of odontoblast cell line MDPC-23 in such membranes. Methods Asymmetric porous chitosan membranes were fabricated successively via the formation of a dense chitosan film by casting method and a porous chitosan sponge layer by lyophilization. Micromorphology of the membranes was examined using scanning electron microscope (SEM). The MS-TGFs were incorporated into the membranes and then MDPC-23 was seeded in them and incubated in vitro. Two days later, biocompatibility was observed with SEM, and proliferation profile was drawn according to the results of cell counting within 7 days. Results Asymmetric chitosan membrane was a bilayer structure, including dense film and porous sponge layer with pores of 60 μm-160μm in diameter. Porosity of the membranes was 83.26%. The ability of banding water was 15.6g water/g membrane. MS-TGFs were seeded into the pores of membranes and well distributed. MDPC-23 could grow well in an extended shape on the pore walls of MS-TGF -incorporated membranes. The membranes containing MS-TGFs significantly augmented the cell proliferation. After 7 days'cultivation, the number of cell increased from (2. 03 ±0. 04) × 10^4 to( 18. 31 ±0. 60) × 10^4. Conclusion Asymmetric porous chitosan membranes were characterized by desired physical properties. After MS-TGFs were seeded into membranes, controlled release of TGF-β1 would be conveyed to cells within the membranes and enhance cell proliferation.
