摘  要:目的:探讨不同浓度MnC l2对树突状细胞(DC)调节T细胞抗肿瘤活性的影响。方法:采用析因实验设计,分析MnC l2浓度、DC细胞浓度、效靶比三因素各水平对DC调节T细胞抗人肝癌细胞株(Huh7)活性的影响;流式细胞仪检测MnC l2最佳刺激组与空白组DC膜分子HLA-DR表达;体内实验分析MnC l2最佳刺激组DC在SC ID小鼠体内对T细胞抗Huh7活性的调节作用。结果:MnC l2浓度、DC细胞浓度、效靶比三因素各水平对T细胞杀伤活性皆产生显著效应(P<0.01),并且3个因素中两两有协同作用(P<0.01)。最佳MnC l2刺激浓度为300μmol.L-1,该浓度MnC l2能显著提高DC表面HLA-DR表达水平及阳性细胞率(P<0.01)。在SC ID小鼠体内,MnC l2刺激组DC成瘤率为2/5,对照组成瘤率为5/5;MnC l2刺激组的肿瘤平均体积为0.42 cm3,对照组为2.95 cm3。刺激组的成瘤率及肿瘤平均体积皆小于对照组(P<0.01)。结论:300μmol.L-1MnC l2能显著提高DC表面HLA-DR表达水平及阳性细胞率;MnC l2激活的DC在体外能显著增强T细胞杀伤活性并在SC ID小鼠体内能显著抑制肿瘤生长。Objective To investigate the effect of MnCl2 (0,150,300 μmol · L^-1)on the antitumor activity of human peripheral blood dendritic cells(DC). Methods Factorial design and orthogonal design were adopted to analysis the effect of MnCl2 (0,150,300 μmol · L^-1 ), DC(2 × 10^3, 1 × 10^4 ,2 × 10^4ml^ -1 ) ,E/T(5: 1,10: 1,20: 1 ) on the anfitumor activity of DC ; flow cytometry was applied to examine the expression of HLA-DR on the groups of MnCl2-stimulated DC and the non- MnCl2-stimulated DC. The effect of the best stimulated -DC by the MnCl2 (300 μmol · L^-1 ) on the cytotoxicity of T cells in SCID micet attacked by hepatic tumor cells (Huh7 )was studied. Results These three factors all had a remarkable effect on the antitumor activity of DC. Both the expression of HLA-DR and the positive percentage of HLA- DR of MnCl2-stimulated DC were much higher than that of non-MnCl2-stimulated DC. The occurrence rate and the general volume of tumor in the SCID group injected with MnCl2-stimulated DC were remarkably less than that of the other. group injected with non- MnCl2-stimulated DC. Conclusion MnCl2 can remarkably enhance the expression and positive percentage of HLA- DR in DC in vitro. MnCl2-stimulated DC can remarkably augment the cytotoxicity of T cells and remarkably suppress the growth of tumor( Huh7 ) in SCID.
