摘 要:目的运用基因芯片技术研究S-生物丙烯菊酯对正常人淋巴细胞基因表达谱的变化。方法取3名正常人外周血淋巴细胞,加入S-生物丙烯菊酯刺激后提取总RNA,合成cRNA并分别用Cy3和Cy5标记,与AgilentHum an1B寡核苷酸基因芯片杂交,研究基因表达谱变化。结果正常淋巴细胞在S-生物丙烯菊酯刺激后共有346个基因表达发生变化,其中16个免疫和防御相关基因表达下调。结论S-生物丙烯菊酯可导致正常淋巴细胞基因表达谱改变,某些免疫和防御相关基因表达下调。Objective To study the effect of S-bioallethrin on the gene expression profile of human lymphocytes. Methods Total RNA were extracted and purified from peripheral blood lymphocytes treated or not treated by S-bioallethrin. cRNA were synthesized and labeled with Cy3 and Cy5 respectively. The labeled cRNA fragments were hybridize with the Agilent human 1B 60 mer oligonucleotide microarray and then the changes of gene expression profile were analyzed. Results changes of expression in 346 genes were found in human lymphocytes treated with S-bioallethrin, 16 genes related to immunity and defense were downregulated. Conclusion S-bioallethrin could induce changes of gene expression of human lymphocytes, and certain genes related to immunity and defense were down-regulated.
