摘 要:目的观察氢醌(HQ)对人胚肺成纤维(HLF)细胞DNA损伤和微核形成情况。方法分别用不同剂量的HQ处理HLF细胞1h,继续培养24h后进行微核试验;处理HLF细胞2h后直接用彗星试验检测DNA损伤。结果各剂量组的微核率为2‰~9‰(P〈0.05),核异常率为3‰~13‰(P〈0.05)。相关分析表明,存在明显的剂量——效应关系,其中20、40和80μmol/L剂量组的微核率和核异常率显著增加(P〈0.05)。各剂量组细胞DNA拖尾率和彗星尾长均有明显的剂量—效应关系,其中20~80μmol/L剂量组的拖尾率和彗星尾长明显增加(P〈0.05);且随着HQ剂量的增加,高度、重度DNA损伤的比例也逐渐增加。结论似乎HQ可致HLF细胞DNA损伤和细胞微核形成。Objective To observe DNA damage and micronucleus formation in human embryo lung fibroblasts (HLF) treated with hydroquinone (HQ). Methods HLF were treated at the doses of 0 ~ 80μmol/L HQ respectively for 1 h and micronucleus test was performed after they were continually cultured for 24 hours, HLF were also treated at the same doses of HQ for 2 hours and DNA damage was detected by comet assay. Results Micronucleus test showed that formation of micronuclei and abnormal nuclei were seen in all groups treated by different doses of HQ, their rates being 2‰ to 9‰(P 〈 0. 05) and 3‰ to 13‰ (P 〈 0. 05) respectively. There were significant increases in rates of micronuclei and abnormal nuclei in cells treated at the doses of 20 - 80 μmol/L of HQ( P 〈 0. 05). In the comet assay, there were Obvious dose-effect relationships in the indexes of percentage of cells with tails and mean tail length in each group treated with different doses of HQ. Very significant increases in percentage of cells with tails and length of their comet tail were observed in those groups treated at the doses of 20 - 80 μmol/L of HQ. And proportions of high and severe DNA damage increased with the doses of HQ increased. Conclusion It was seemed that HQ could induce DNA damage and micronucleus formation in human embryo lung fibroblasts.
