摘 要:目的:探讨环境雌激素(EEs)对雄性胎鼠睾丸下降以及睾丸引带肌动蛋白表达的影响。方法:成熟雌性昆明小鼠140只随机分为14组,孕9~17d每天分别给予己烯雌酚、17α雌二醇和17β雌二醇[每种药物分别给予不同剂量25、50、100、200μg/(kg·d)],对照组、正常组分别给予等体积的二甲基亚砜、NS。孕17d处死母鼠后观察雄性胎鼠睾丸下降情况,应用透射电镜观察睾丸引带细胞超微结构,应用免疫组化检测睾丸引带组织中肌动蛋白的表达。结果:EEs可致睾丸发育较差,下降不全,与对照组比差异有统计学意义(P<0.05)。透射电镜观察发现正常组及对照组睾丸引带细胞内肌丝粗大,密体明显,胞浆中细胞器丰富;实验组睾丸引带细胞内肌丝明显细小,密体不明显,胞浆中细胞器较少,随EEs剂量增大此表现更明显。实验组肌动蛋白含量降低,EEs剂量越大,肌动蛋白含量越低(P<0.05),有显著的剂量效应关系。结论:孕期应用EEs可影响雄性胎鼠睾丸下降、睾丸引带细胞超微结构以及肌动蛋白的表达,这些改变可能是EEs导致多种生殖系统畸形的直接或间接原因。Objective To explore the effects of environmental estrogens(EEs)on testicular descent and expression of actin in gubemaculum testis (GT) of fetal male mice. Methods One hundred and forty pregnant Kunming female mice were randomly assigned into 14 groups( n=10 for each group). DES, 17α-E2 and 17β-E2 were subcutaneously administered at a dose of 25, 50, 100,200 μg/(kg·d) from gestational day(GD)9 through GD17. The control group and normal control group were given dimethyl sulfoxide(DMSO) and saline alone, respectively. They were sacrificed on GD17. The position of testes was recorded, ultrastructural changes of GT were observed under transmission electron microscopes(TEM) . The expression intensity of aetin in GT was quantitated by immunohistochemistry. Results EEs could affect the testicular descent, the testis position was higher than that in control and normal control groups( P〈0.05) . Some smaller disordered myofibrils and sparse cytoplasmic organelles in the gubernacular muscular cells were observed in the treated groups. The expression intensity of actin in GT was significandy weaker than that in control and normal control groups(P〈0.05) . Conclusion Prenatal exposure to EEs can affect the testicular descent, the uhrastructural changes and the expression of actin in GT. These changes maybe the direct or indirect causes of variant kinds of genital malformation such as cryptorchidism and hypospadias.
