摘 要:目的观察基础状态下成年高血压大鼠(SHR)心肌成纤维细胞(CFs)的吸光值、增殖细胞核抗原(PCNA)表达、3H-胸腺嘧啶核苷(3H-TdR)及3H-脯氨酸掺入法(3H-Proline)掺入量和NO含量的变化。方法胰酶消化法分离、培养大鼠CFs,分别用改良的MTT比色法、免疫组化法3、H-TdR掺入法观察CFs的增殖状况,用3H-Proline观察CFs的胶原合成情况,用硝酸还原酶法检测CFs培养液中的NO含量。结果培养72 h,SHR组与正常血压组比较,CFs吸光值、PCNA表达3、H-TdR及3H-Proline掺入量增高具有非常显著意义(P<0.01),NO含量降低也具有非常显著意义(P<0.01)。结论CFs在维持基质间隙中起着关键作用,其增殖、胶原合成以及NO含量的异常都可以导致左心室肥厚的产生和发展。AIM To evaluate the changes of MTT OD value,PCNA protein expression ,^3H-TdR and ^3H- proline incorporation and NO content in rat cardiac fibroblasts(CFs) derived from SHR. METHODS CFs were isolated by trypsin digestion method. Modified MTT array was used to determine cell number.^3H-TdR incorporation was used to analyze PCNA protein expression and determine DNA synthesis. Collagen synthesis and NO content were also detected. RESULTS Under basal condition (72 h), the A value, PCNA protein expression,^3H-TdR and ^3H-proline incorporation derived from SHR were significantly greater than those of Wistar rats (P 〈0.01 ), while NO content derived from SHR were significantly lower than that of Wistar rats ( P 〈 0.01 ). CONCLUSION The abnormal changes of CFs, collagen content of CFs and NO content in SHR might be related to the formation and development of left ventricular hypertrophy.
