摘 要:目的观察缺氧大鼠心肺组织中新发现的小分子活性肽apelin的变化,探讨其在缺氧性肺动脉高压发生发展中的病理生理学意义.方法雄性SD大鼠20只,分为对照组和缺氧组(9%~11% O2,9 h/d,6 d).导管法测定平均肺动脉压(mPAP)、平均颈动脉压(mCAP),称重法测定右心室与左心室加室间隔重量比(RV/LV+S).放免法测定血浆、右室心肌及肺组织匀浆apelin水平,RT-PCR检测右室心肌及肺组织apelin mRNA表达的变化.结果①缺氧组大鼠mPAP较对照组高26.06%(P〈0.05),RV/LV+S较对照组高10.83%(P〈0.01).mCAP两组间差异无统计学意义.②缺氧组大鼠右心室肌、肺组织apelin浓度(pg/mg)较对照组分别低16.33%(P〈0.05)和14.10%(P〈0.01),血浆apelin浓度(pg/ml)两组间差异无统计学意义.③缺氧组大鼠心肌和肺组织apelin mRNA 表达水平较对照组分别低22.95%和22.82%(均P〈0.05). 结论缺氧大鼠右室心肌及肺组织局部apelin蛋白和基因表达的显著下降,可能是导致大鼠缺氧性肺动脉高压发生、发展的重要因素之一.Objective To investigate the probable function of new peptide apelin in pathological proceeding of pulmonary hypertension induced by hypoxia. Methods Twenty male Sprague-Dawley rats were randomly divided into normal control group and hypoxia group (9% - 11% 02,9 h/d, 6 d). The mean of pulmonary arterial pressure ( mPAP), mean carotid arterial pressure (mCAP) and the weight ratio of fight ventricle to left ventricle plus septum (RV/LV + S) were measured. The concentration of apelin in the plasma, fight ventricular myocardium and lung tissue were measured by radioimmunity. The expression of apelin gene in right ventricular myocardium and lung tissue were measured by reverse transcription-polymerase chain raction (RT-PCR). Results The mPAP and RV/( LV + S) of hypoxic group were respectively more by 26.06% and 10.83% than control group ( P 〈 0.05, P 〈 0.01 ), while mCAP was of no significant difference between the two groups. The apelin concentration (pg/mg) of right ventricular myocardium and lung tissue of hypoxic group were lower by 16.33% and 14.10% than control group (P 〈0.05, P 〈0.01 ), but the plasma level of apelin (pg/ml) was of no difference. The expression of apelin mRNA in right ventricular myocardium and lung tissue of hypoxic group were lower by 22.95 % and 22.82% than control group ( P 〈 0.05 ). Conclusion The local decreased expression of apelin gene and peptide in the right ventricle and lung tissue in rats might be an important factor in the development of pulmonary hypertension induced by hypoxia.
