摘  要:目的通过对RIM3γ mRNA在中枢神经系统和抑制性神经元中的特异性表达的研究,探讨RIM3γ的生理功能.方法使用成年Wistar大鼠,通过原位杂交技术和双标免疫荧光检测法对RIM3γ mRNA在中枢神经系统和抑制性神经元中的特异性表达进行研究.结果RIM3γ的mRNA主要表达在丘脑腹后外侧核、嗅球的僧帽细胞层、大脑皮质躯体感觉区的第Ⅱ~Ⅳ层及在小脑的颗粒层和脊髓后角的边缘层区域.双标免疫荧光检测法显示RIM3γ分布在大鼠的大脑皮层、脊髓和小脑的某些抑制性神经元. 结论RIM3γ在大鼠中枢神经系统的特异性表达,为进一步探索RIM3γ的生理功能提供了基本依据.Objective By mapping specific expression of RIM3γ mRNA in the central nervous system and inhibitory neurons, we probed into physiological function of RIM3γ. Methods Adult Wistar rats were used in the present study. The brain and cervical enlargement of the spinal cords of the adult Wistar rats were dissected. Using In situ hybridization and double labeled fluorescence assay, we demonstrated the specific expression of RIM3γ mRNA in the central nervous system and inhibitory neurons. Results RIM3γ mRNA was preferentially expressed in lateral posterior thalamic, mitral layer of the main olfactory bulb, layers Ⅱ-Ⅳ of somatosensory cortex, the granula layer of the cerebellum and the superficial layers of the spinal cord dorsal horn. Double labeled fluorescence assay showed the expression of RIM3γ in some inhibitory neurons of the cortex, the spinal cord and the cerebellum of adult Wistar rats. Conclusion Specific expression of RIM3γ mRNA in the CNS of rats provides the basis for further exploring physiological function of RIM3γ.
