曲尼司特滴眼液对兔眼Tenon囊组织创伤愈合反应的影响

摘　　要：目的活体观察0.5%曲尼司特滴眼液对兔眼Tenon囊组织α2(Ⅰ)链前胶原及转化生长因子-β1(TGF-β1)mRNA表达的影响。方法15只新西兰兔随机分为3个组:实验组(应用0.5%曲尼司特滴眼液)、溶媒对照组(应用基础溶液)、正常对照组。采用半定量逆转录聚合酶链反应(RT-PCR)观察曲尼司特滴眼液对兔眼Tenon囊组织α2(Ⅰ)链前胶原及TGF-β1mRNA表达的影响。结果实验组、溶媒对照组、正常对照组的α2(Ⅰ)链前胶原/β-actin象素值比值分别为0.880±0.029、1.034±0.065、0.808±0.041,各组比值差异有显著统计学意义(q=4.808,3.207,6.551,P<0.05)。3个组的TGF-β1/G3PDH象素值比值分别为0.894±0.058、0.882±0.090、0.852±0.061,实验组与溶媒对照组的比值虽高于正常对照组,但各组差异无显著统计学意义(F=0.463,P>0.05)。结论0.5%曲尼司特滴眼液能明显抑制兔眼Tenon囊组织α2(Ⅰ)链前胶原mRNA的表达,可作为抗纤维化药物用于青光眼非穿透性小梁切除术后。Objective Tranilast has been determined to inhibit fibroblast proliferation and collagen synthesis in vitro. Topical use of tranilast following trabeculectomy can enlarge the filtering bleb. The present study in vivo was designed to investigate the effect of 0.5% tranilast eye drops on alpha 2 （ Ⅰ ）procollagen and transforming growth factor-β1 （TGF-β1 ）mRNA expression in rabbit Tenon＇ s capsule. Methods Fifteen rabbits were randomly divided into tranilast group, vehicle solution group and control group. Tranilast eye drops were topically used for 4 times daily in tranilast group, and vehicle solution was used at the same method in vehicle solution group and no any eye drops was used in control group. Alpha 2 （ β ） procollagen and TGF- β mRNA in rabbit Tenon ＇ s capsule were detected by reverse transcriptase - polymerase chain reaction （ RT-PCR ） analysis. Results The value of alpha 2（ Ⅰ） procollagen/β-actin was 0.880 ±0.029,1.034 ±0.065,0.808 ±0.041 in tranilast group, vehicle solution group and control group respectively and was the statistically different among the three groups（ q = 4. 808,3. 207, 6. 551, P 〈 0. 05 ）. The value of TGF-β1/G3PDH was 0. 894 ± 0. 058,0. 882 ± 0. 090,0. 852 ± 0. 061 in tranilast group, vehicle solution group and control group respectively, showing a insignificant difference among them （ F = 0. 463, P 〉 0. 05 ）. Conclusion 0. 5 % tranilast eye drops could inhibit expression of alpha 2 （ Ⅰ ） procollagen mRNA in rabbit Tenon＇ s capsule. It can be used as an anti-scarring drug after non-penetrating trabecular surgery.
