用固相萃取、GC／NPD技术分析全血及脏器中5种三环类抗抑郁药物

摘　　要：本文介绍了用国产ＧＤＸ－４０３固相柱富集全血和肝组织中的阿米替林、三甲丙咪嗪、丙咪嗪、多虑平、氯丙咪嗪等５种三环类抗抑郁药物（ＴＣＡｓ），以ＧＣ／ＮＰＤ毛细管色谱技术进行分析的方法。ＳＫＦ_（５２５Ａ）作内标。方法简单、快速。１ｍｌ全血中加入上述药物２．０μｇ回收率为８３．６％～８９．７％，ＲＳＤ为２．４３％～６．４４％；１ｇ肝中加入药物４．０μｇ，回收率为６２．０％～８７．７％，ＲＳＤ为２．７５％～７．０３％。血中检出限为５～３０ｎｇ／ｍｌ。方法经动物实验验证，结果满意。A simple and rapid method for the isolation of five tricyclic antidepressant drugs:amitriptyline, trimipramine, imipramine, doxepin. clomipramine with GDX_(403) column from whole blood and liver samples is presented. SKF525A was used as the internal standard. samples of blood and hydrolysis-digested liver were collected and purified by SPE column, the column eluate was analyzed by using GC/NPD. Recovery studies were performed by adding 2. 0μg of drugs to 1ml of whole blood and 4.0μg of drugs to lg liver samples. The recoveries of fivedrugs from blood samples ranged from 83. 6% to 89. 7% with RSD less than 6.44% and liver samples ranged from 62% to 87. 7% with RSD less than 7% , respectively. The detection limit for blood samples was 5～30ng/ml. The procedure is presently being used in case work.
