摘 要:本文观察了缺锌对大鼠股四头肌深层肌总蛋白质、DNA、总RNA、肌动蛋白(DNaseⅠ抑制法)和α-肌动蛋白mRNA(地高辛标记探针,Northern杂交)含量的影响。结果显示:缺锌时肌肉中总蛋白质、DNA和肌动蛋白的相对含量变化不明显,总RNA和α-肌动蛋白mRNA的含量明显减少。这提示,锌缺乏导致α-肌动蛋白的基因表达下降,可能在转录和翻译过程中影响骨骼肌肌动蛋白的合成速率。he concentrations of zinc,total protein,DNA,total RNA,actin(by DnaseⅠassay),and α一actin mRNA(by dikroxigenin- labeled cDNA probe,Northern blot) were deterinined in the profouiid muscularis of m,quadriceps fenioris of niJe Sprague-Dawley rats with different zinc status: controlled rats fed with controlled diet(Zn 50mg/kg diet)for 3 weeks,zinc一deficient(ZnD) rats fed witli ZnD diet (Zn 1mg/kg diet)for 3 weeks,and zinc- reple-nished (ZnR) rats,wtih ZnD diet for 2 week5,and then controlled diet for 1 week.The results showed that the contents of zinc,total protein,DNA and actiiis iii the muscles of 3 groups of rats were not significantly different.The total RNA and α一actin mRNA decreased significantly in ZnD rats, and increasedagain inthe ZnR rats.It was suggested that the decr-eased a,actin gene expression by zinc deficiency may influence the synthe-tic speed of actin drt transcription and traiislation levels in skeletal inuscle.
