摘  要:目的:建立野葛根总黄酮提取及分离工艺。方法:采用正交试验考察乙醇浓度、乙醇用量、提取时间、提取次数4个因素及大孔吸附树脂分离技术对野葛根总黄酮得率的影响。结果:野葛根的适宜提取工艺为用8倍量60%乙醇,提取3次,每次1.5h,洗脱液为70%乙醇。结论:该工艺葛根总黄酮得率约为8%,以葛根素计含量为60%以上,适宜工业化生产。OBJECTIVE To design the extraction and separation technology of the total flavonoids in Radix Pueraria lobata. METHODS The orthogonal-test method was adopted to research the effect of the 4 factors,which include the concentration and volume of alcohol,the time and times of extraction, and the technique of adsorption separation through macroporous resin column on the yielding rate of total flavonoids in Radix Pueraria lobata. RESULTS The optimum extraction conditions were: extracting three times with 60%alcohol 1.5 h each time with 8 times the voume of the alcohol. The eluent was 70% alcohol.CONCLUSION The yielding rate of the general flavonoids is about eight percent by using the technology. The content of Puerarin accounts for more thin 60 percent in extractive,so this technology is suitable for industrial production.
