摘  要:目的:建立测定大鼠血浆中天麻素含量的方法。方法:取出注射天麻提取物后大鼠的血液,用无水乙醇沉淀血浆中的蛋白质并把醇溶液用氮气吹干,加注射用水溶解后直接用高效液相色谱法测定。色谱柱:YMC-Pack ODS(250mm×4.6mm,5μm);流动相:乙腈-0.05%磷酸(3:97,v/v);流速:1.0mL·min^-1;检测波长:220nm;柱温:室温。结果:天麻素最低血浆检测浓度为5mg·L^-1,线性范围为0.1~1.0μg,r=0.9992。平均加样回收率为98.59%,RSD为1.71%(n=6)。经大鼠血液循环后,天麻素供试品图谱主峰单一。结论:该法简便易行,可以用于测定血浆中天麻素的含量。OBJECTIVE To set up a pretreatment method of plasma sample of rat after injecting extraction of Gastrodia elata BL and the determination of the content of Gastrodin in plasma by HPLC. METHODS After the rats were injected with the extraction, the plasma samples were drawn and treated by adding ethanol and the supernatant was dried by N2. The residue was redissolved in water,then detected by HPLC directly. The analysis was carried on an analytical column YMC-Pack ODS (250 mm× 4. 6mm,5μm). The mobile phase consisted of acetonitrile -0. 05 % H3 PO4 ( 3:97, v/v) and the flow rate was 1.0 mL· min^- 1. The UV detection wave length was at 220 nm and the column was kept at room temperature. RESULTS The detection limit was 5 g·L^-1, The linear relationship was within the range of 0. 1 - 1.0μg(r = 0. 999 2). The average recovery was (98. 60± 1.68) % (RSD= 1.70% ,n = 6). CONCLUSION The method is shown to be suitable for determination of Gastrodin in plasma.
