摘 要:本文探讨了持续性运动和/或注射脱氢表雄酮(DHEA)对雄性大鼠体重增长、血脂水平、以及肝脏、心脏及腓肠肌细胞内过氧化物体增殖活化受体α(PPARα)及其靶酶-酰基辅酶A氧化酶(AOX)的mRNA和PPARα蛋白量的影响。将40只SD大鼠随机分为4组:A为对照组,B为DHEA组,C为运动训练组,D为运动训练+DHEA组。DHEA给药方式:每天皮下注射20m g/kg的DHEA S,每周6d,共6周。运动方式:采用跑台,速度为25m/m in,每天1h,每周6d,共7周。结果发现:(1)D组实验后体重增长明显少于A组和B组(p<0.05)。(2)各组之间的总胆固醇、甘油三酯、高密度脂蛋白胆固醇和低密度脂蛋白胆固醇含量均无明显差异。(3)各组间的PPARα和AOX的mRNA表达均无明显差异。(4)在肝脏,C组和D组的PPARα蛋白水平明显高于A组(P<0.01和P<0.001)。在心脏,B组、C组和D组的PPARα蛋白水平明显高于A组(P<0.05、P<0.001和P<0.001)。在腓肠肌,D组的PPARα蛋白水平明显高于A组(P<0.05)。结果表明:(1)运动训练加注射DHEA可以明显控制体重的增长。这种作用可能不是通过降低血脂水平而实现的。(2)DHEA和持续性运动在翻译及翻译后水平上调了PPARα基因的表达。DHEA提高心脏内PPARα的水平,而持续性运动能诱导心脏和肝脏内PPARα的表达。DHEA结合运动训练诱导肝脏、心脏和腓肠肌的PPARα的表达,介导各脏器的脂肪分解。The purpose of this study is to discuss the effects of dehydroepiandrosterone (DHEA) and/or endurance training on the body weight,blood lipids ,the expression of peroxisome proliferator-activated receptor α (PPARα) and acyl-CoA oxidase (AOX) in some tissues in rats. Forty adult male Sprague-Dawley rats were randomly divided into four groups:group A (control) , group B (DHEA),group C (training) and group D (DHEA and training). DHEAS was administrated by subcutaneous injection at a daily dose of 20mg/kg body weight in group B and D for six weeks. Animals in group C and D received endurance training by running on the treadmill for one hour at a speed of 25m/ min,six days per week,for seven weeks. The results showed that: (1) The increased body weight in group D was less than in group A (P〈0. 05) and B (P〈0. 05) significantly. (2) There were no significant differences in serum concentration of total cholesterol, triglyceride, HDL C and LDL-C in all groups. (3) There were no significant differences in mRNA expression levels of PPARα and its target gene acyl-CoA oxidase (AOX) in liver,heart and gastrocnemius muscle in all groups. (4) The level of PPARα protein was significantly higher in group C (P〈0. 01) and D (P〈 0. 001) than that in the control group in liver. The level of PPARα protein was significantly higher in group B (P〈0. 05) ,C (P〈0. 001) and D (P〈0. 001) than in the control group in heart. The level of PPARα protein was significantly higher in group D (P〈0.05) than in the control group in gastrocnemius muscle. The results also indicated that: (1) DHEA combined with endurance training could reduce the weight gain. This effect might not be related to the blood lipids level. (2) DHEA and endurance training up-regulated the expression of PPARα gene in translation and post-translation level. DHEA led to the higher level of PPARα protein in heart,endurance training led to the higher level of PPARα
