摘  要:目的 探讨广州地区儿童感染的星状病毒ORF2基因特点和基因类型。方法 参考CenBank上的星状病毒Ⅰ型ORF2基因设计了两对特异性引物,进行巢式PCR扩增出目标片段,克隆于T载体上,测定序列,用phylip3.65软件、NJ法构建进化树。结果 星状病毒ORF2基因为2316bp,编码771个氨基酸,ClustalW同源性比较发现,Hastv-gz克隆与4型星状病毒氨基酸同源性为最高(93%),与其它基因型的同源性为61%~70%。结论 广州地区儿童腹泻感染的星状病毒Hastv-gz克隆ORF2基因序列为2316bp,Hastv-gz克隆与4型星状病毒同源性为93%,以phylip3.65软件、NJ法构建ORF2基因进化树中,Hastv-gz与4型星状病毒密切相关,确认Hastv-gz是4型星状病毒。Objective To study the ORF2 gene and genotype of human astrovirus infected infants in Guangzhou of China. Methods Primers were designed on the basis of the ORF2 sequence of genotypel astrovirus in the GenBank, and amplify the astrovirus ORF2 gene by nest PCR. The PCR - products were cloned to T vector and sequenced, phylogenetic tree was constructed by the neighbor - joining method in Phylip3.65. Results The ORF2 gene of Hastv - gz was 2316bp in length, and compared with those of astrovirus in GenBank, revealed that the highest homology is with genotype 4 (93 % ). Homology with other genotypes ranged between 61% and 70%. Conclusions The complete nucleotide sequence of ORF2 gene isolated from Guangzhou in China is 2316bp in length, Comparison of the ORF2 of Hastv - gz with the known sequences of types 1 - 8 show the highest homology is with genotype 4 (93%); phylogenetic tree is constructed by the neighbor-joining method with the Porcine astrovirus(Y15938) as an outgroup, tree shows that Hastv- gz is close to genotype4 astrovirus, and suggests the Hastv - gz clone belongs to genotype (serotype) 4 astrovirus.
