摘 要:目的:探讨中华眼镜蛇毒(CCV)对大鼠肾缺血/再灌注(I/R)损伤的保护作用及其机制。方法:雄性SD大鼠55只,随机分为实验组(25只)、对照组(25只)和假手术组(5只)。实验组与对照组分别腹腔注射0.1%的CCV(0.4mg/kg)和1ml的生理盐水,12h后建立肾I/R损伤模型,按处死时间不同又分为5个亚组,每组5只;假手术组单纯分离双肾动脉。按设计时间点留取血清。观察各组血肌酐(Scr)和尿素氮(BUN)、髓过氧化物酶(MPO)、补体C3和C4的变化。结果:肾I/R损伤后,对照组Scr和BUN显著升高,再灌注12h后达正常值5倍以上,MPO水平上调,补体C3水平先升高后降低,1h组与假手术组相比有统计学差异(P<0.05),补体C4水平变化不大。经CCV预处理的实验组,Scr、BUN和MPO值明显下降,补体C3水平低于假手术组,C4水平变化不大。结论:CCV预处理可明显减少补体C3和MPO的活化,改善肾功能。Objective:To investigate the renal protection and mechanism of ChinesCobra Venom(CCV )in renal isehemia/reperfusion injury rat models. Methods.55 rats were randomly divided into three groups; CCV group(25 eases), control group(25 eases) and sham operation group(5 cases).0.1% CCV(0, 4 mg/kg)and 1 ml saline solution was infused into abdominal cavity 12 h before ischema in CCV group and control group respectively. CCV group and control group were randomly divided into 5 teams according to the time when the rats are sacrificed, Sham operation group only separated double renal arteries. Sample of serum acquired on the basis of different time. Renal function was measured by BUN and Scr, Complement c3 and c4 was observed. Neutrophil infiltration was assessed by myeloperoxidase(MPO), Results:BUN,Set and MPO activation rised gradually and reached peak after 12 h reperfusion in control group. Complement C3 increased at the beginning and decreased in the end, there is obvious difference only between lh group and sham operation group (P 〈 0.05). Serum Bun/Scr and MPO activaty decreased significantly in CCV group than control group and meanwhile .serum C3 was lower than sham operation group. However, There was no difference of C4 between CCV group and control group, Conclusion: Pretreating the rats with ccv can reduce C3 and MPO activation and improve renal function.
