摘  要:目的建立中毒检材的毒鼠强急性毒性实验方法和毒鼠强GC/MS法快速检验方法快速诊断毒鼠强中毒.方法中毒检材大鼠或小白鼠灌胃,观察反应.固体检材匀浆或液体检材,加入内标SKF525或地芬尼多(眩晕停),硼砂缓冲液碱化(pH=9.18),苯萃取2次,氮气挥干,无水乙醇定容, GC/MS检测,质谱特征碎片定性分析,选择离子监测模式(SIM),校正曲线法定量分析.结果毒鼠强灌胃后大鼠出现特有强直性抽搐.毒鼠强的质谱特征碎片为:212和240. GC/MS检测毒鼠强的线性范围为(0.05~4) μg/mL,最低检出浓度为0.02 μg/mL.2002年-2005年检测的74例毒鼠强中毒死亡案件中,56例可疑毒鼠强中毒者中18例检出毒鼠强,其中31例可进行大鼠毒性实验者中10例大鼠毒鼠强毒性实验阳性,185例可疑毒鼠强中毒死亡者中,74例检出毒鼠强,其中30例可进行大鼠毒性实验者中15例大鼠毒鼠强毒性实验阳性.结论动物实验筛选、GC/MS法快速诊断毒鼠强中毒,具有快速、准确、操作简便,结果可靠,可应用于毒鼠强中毒的诊断和毒鼠强中毒死亡的法医学鉴定.Objective To develop an animal experiment screening and gas chromatography with mass spectrometry identification for rapid diagnosis of tetramine poisoning. Methods Rats was treated by intragastric administration of tetramine samples. After internal standard SKF525 or diphenidol were added, the solid sample homogenate or liquid sample was alkalified with a borax buffer (pH= 9.18) and extracted with benzene twice. The qualitative and quantitative analysis of tetramine was carried out by gas chromatography with mass spectrometry identification with a SIM model. Results The rats showed a special and spastic convulsion following intragastric administration of tetramine poisoning samples. The mass - spectrum characteristic fragments of tetramine were 212 and 240. The extraction curves of tetramine in serum was linear over the range of 0.05 μg/mL to 4 μg/mL, The limit of detection was 0.01μg/mL. Tetramine were identified in 18 out of 30 doubtful tetramine poisoning, And 10 out of 15 tetramine poisoning cases showed a positive animal experiment. Tetramine were identified in 74 out of 185 with doubtful tetramine poisoning. And 31 out of 56 tetramine poisoning death cases showed a positive animal experiment. Conclusion The animal experiment screening and GC/MS analysis for rapid diagnosis of tetramine poisoning was a simpler, rapider, more sensitive and more practiced method. It was good for diagnosis of tetramine poisoning and identification of tetramine poisoning death.
