摘 要:目的探讨空肠弯曲菌(Campylobacter jejun i,CJ)亚单位脂质体佐剂疫苗免疫新西兰兔后Th1/Th2应答机制。方法制备CJ亚单位阳离子和阴离子脂质体佐剂疫苗,用不同剂量(按蛋白质含量计算,0.25mg/kg、0.5mg/kg、0.75mg/kg)的疫苗免疫新西兰兔,于末次免疫后10d采集血清,ELISA(双抗体夹心法)检测各组动物血清中IL-2、IFN-γ、TNF-β、IL-4、IL-6、IL-10的水平,同时设有CJ完全福氏佐剂免疫原对照组和生理盐水(NS)对照组。结果免疫组与CJ完全福氏佐剂免疫原对照组和NS对照组相比较,IL-4、IL-10水平与对照组相比有显著性升高(P<0.01),其中同一剂量(0.5mg/kg)不同佐剂组以CJ亚单位阳离子脂质体佐剂疫苗组升高明显(P<0.05),不同剂量同一佐剂疫苗组以0.5mg/kg组升高较明显(P<0.01),IL-2、IFNγ-、TNF-β水平各免疫组与对照组相比无显著性差异(P>0.05)。结论CJ亚单位脂质体佐剂疫苗诱导了以Th2应答为主的免疫应答类型。Objective The present study wits designed to explore the immune mechanism of Thl/Th2 response in New Zcaland white rabbits immunizcd witl, subunit liposomal adjuvant vaccine of Campylobactcr jejuni (CJ). Methods The subunit vaccines of CJ were prepared with cationic liposome(CL), antionic liposome(AL) as adjuvants , rabbits were immunized by different dosages( measured by protein conte,0. 25mg/kg,0.5mg/kg,0.75mg/kg) of subunit adjuvant vaccine of CJ. On 10th day after the final immunization, all mbbits were saerificed and their sera were collected, The levels ofcytokine IL-2 ,IFN-γ ,TNF-β, IL-4, IL-6 and IL-10 in serum were tested with sandwich enzyme-linked immunosorbert assay (ELISA), respectively, rabbits immunized with the CFA immunogen and physiological saline solution (NS) were used as controls. Results Immunized group compared with the Complete Freund's Adjuvant (CFA) immunogen and the control group. Statisfically increased levels of serum eytokine IL-4, IL-10 were observed in immunized rabbits with liposomal adju- vant vaccine groups compared with thc NS control group ( P 〈 0.01 ) , the level of positive liposomal adjuvant group increascd more apparcutly under the same dosage with other groups ( p 〈 0.01 ) , contrarily the 0.5mg/kg group had the higest serum level under the same adjuvant ( P 〈0.01 ) ,no stastisfic diffcrence were measured in the serum levels of IL-2, IFN-γ and TNF-β between immunizcd group and eonlrel group ( P 〉 0.05 ). Conclusions The results inelicated the subunit liposonmal adjuvant vaccine of CJ can induce Th2 type of immune response.
