摘  要:目的:观察虫草制剂对进行性肾小球硬化大鼠细胞外基质的影响,并探讨其作用机制。方法:实验于2002-11/2003-11在青岛大学医学院中心实验室完成。将36只健康级Wistar大鼠随机数字法分为正常对照组、模型组和冬虫夏草组,每组12只。模型组和冬虫夏草组分别行5/6肾切除,制备进行性肾小球硬化模型。正常对照组行两次手术,但不切除肾脏。冬虫夏草组在术后3d给予四川产天然冬虫夏草制剂百令胶囊4g/(kg·d)水提取液灌胃进行干预治疗。末次术后4,9周分别测定大鼠血生化;观察肾脏病理变化,用免疫组织化学方法检测肾组织胶原Ⅳ及纤维连接蛋白。结果:36只大鼠纳入结果分析。①虫草制剂组能够减少尿蛋白排泄(25.03±3.36)g/L,降低血胆固醇(1.38±0.21)mmol/L和低密度脂蛋白(0.86±0.11)mmol/L,与模型组相比差异显著(P<0.01)。②虫草制剂组术后9周时肾小球硬化指数为30.33±7.74,与模型组44.67±9.87相比下降明显,胶原Ⅳ和纤维连接蛋白表达也减少(P<0.05~0.01)。③虫草制剂组在术后4周基质金属蛋白酶组织抑制因子-2mRNA表达减少为(1.668±0.355)A;术后9周基质金属蛋白酶-2mRNA表达增加为(0.643±0.136)A,而基质金属蛋白酶组织抑制因子-2mRNA表达减少为(1.563±0.307)A(P<0.05~0.01)。结论:虫草制剂能够减少5/6肾切除大鼠尿蛋白排泄量;纠正脂质代谢紊乱;调节基质金属蛋白酶/基质金属蛋白酶组织抑制因子系统之间的平衡;减轻肾小球硬化细胞外基质的积聚,延缓肾功能减退。AIM: To investigate the effect of cordycepssinensis on extracellular matrix (ECM) of rats with progressive glomerularselerosis, and probe into its mechanism. METHODS: The experiment was conducted in the Central laboratory of Medical College of Qingdao University between November 2002 and November 2003. A total of 36 healthy Wistar rats were randomly divided into normal control group, model group and cordycepssinensis group with 12 rats in each group. Animals in model group and cordycepssinensis group were respectively nephrectomized for 5/6 to establilsh models of progressive glomerularselesis. Rats in the normal control group received two operations with kidney intact. At 3 days after operation, rats in the cordycepssinensis group received gastric perfusion of water extract 4 g/(kg·d), in which natural cordycepssinensis bailing capsule dissolved. The serum biochemical indicators of rats were determined respectively at 4 and 9 weeks after the last operation, and the renal pathological changes were evaluated. Immunohistochemical method was used to examine the expressions of fibronectin (FN) and collagen Ⅳ in nephridial tissue. RESULTS: A total of 36 rats were involved in the analysis of results.①The urinary albumin excretion in rats of the cordycepssinensis group was reduced (25.03±3.36) g/L, and the blood cholesterol (1.38±0.21) mmol/L as well as low density lipeprotein (LDL) (0.86±0.11) mmol/L were reduced, and the differences in comparison with the model group were remarkable (P 〈 0.01).②At 9 weeks after operation, the glomerular sclerotic indicators in rats of the cordycepssinensis group was 30.33±7.74, which was obviously decreased in comparison with the model group 44.67±9.87, and the expressions of FN and collagen Ⅳ were also reduced (P 〈 0.05-0.01).③The expression of matrix metalloproteinaso inhibiting factor -2mRNA in rats of cordycepssi- nensis group decreased to (1.668±0.355)A at 4 weeks after operation, and the expression of matrix
