摘  要:目的探讨蛋白酪氨酸磷酸酶SHP-1 mRNA及蛋白表达在γ射线体外诱发的白血病小鼠骨髓细胞中的变化情况。方法实验分为白血病组、辐射未癌变组及对照组,应用荧光定量PCR法及W estern b lot法分别检测各组骨髓有核细胞中SHP-1 mRNA及蛋白表达的变化情况。结果白血病组SHP-1 mRNA含量为(5.26±2.14),明显高于辐射未癌变组(3.68±1.27)及对照组(2.95±1.09),(P<0.01);白血病组SHP-1蛋白表达量(光密度)为(5.378±2.905),明显高于对照组(1.852±0.711)及辐射未癌变组(1.559±0.506)。结论在γ射线诱发的白血病小鼠骨髓细胞中,SHP-1 mRNA含量及蛋白表达量均明显增强。Objective To investigate alteration of SHP-1 mRNA and protein of bone marrow cells of leukemia mice after γ-ray exposure. Methods A total of 318 BALB/c mice were exposed to ^60Co γ-ray once a week for 4 weeks, and the total dose of γ-ray received by mice was 7.00 Gy. By pathological examination, 39 mice developed thymic lymphoma, 14 acute lymphocytic leukemia, 21 T-lymphoblast leukemia/lymphoma, 1 spiroma, 4 malignant yolk sac tumor. Exposed to γ-ray, the mice that developed leukemia or were free of canceration were used in our study (n = 10 in each group) , and another 10 mice free from irradiation served as control. SHP-1 mRNA and protein in femoral bone marrow cells were detected by real-time fluorescence quantitative PCR (FQ-PCR) and Western blotting respectively. Results SHP-1 mRNA in leukemia mice was (5.26±2.14), significantly higher than that of mice free of canceration (3.68 ± 1.27) and controls (2.95 ± 1.09) ( P 〈 0.01 ). SHP-1 protein in leukemia mice [ optical density (5. 378 ± 2. 905 ) ] was significantly higher than that in mice free of canceration [ optical density ( 1. 559 ± 0.506 ) ] and controls [ optical density ( 1. 852 ± 0.711 ) ]. Conclusion SHP-1 mRNA and protein were significantly increased in bone marrow cells of leukemia mice induced by γ-ray irradiation.
