溴隐亭对小鼠生育的影响及其作用机理

摘　　要：为研究溴隐亭对小鼠妊娠及免疫系统的影响,于孕5-7d皮下注射不同剂量溴隐亭,孕10d剖杀,统计流产率和平均胚胎数,用酶联免疫吸附法(ELISA)测定血清及子宫匀浆中催乳素、孕酮、干扰素γ(IFNγ)、白介素10(IL10)的含量。结果显示:小鼠流产率随溴隐亭剂量的增加而升高,0.075mg溴隐亭组的流产率为75%,平均胚胎数为2.167±4.064只;0.1mg和0.2mg组均为100%流产,与对照组比较差异显著;随着溴隐亭剂量的增加,血清及子宫组织中IFNγ含量上升,催乳素、孕酮及IL10含量下降。提示溴隐亭具有致小鼠流产的作用,此流产模型与习惯性流产(RSA)都具有偏向Th1型细胞免疫反应的特点,为研究母胎界面的免疫关系提供了实用的动物模型.To investigate the effect of Bromocriptine on the immune system during pregnancy, mice were injected with different dosages of Bromocriptine subcutaneously on days 5 to 7 of pregnancy, and sacrificed by cervical dislocation to calculate abortion rates and the average numbers of fetus. Levels of Prolactin, Progesterone, Interferon gamma （IFN） -γ, Interleukin （IL） -10 in serum and uterus supernatant were measured using Enzyme-Linked Immuno-absorbent Assay （ELISA）. The results showed that abortion rates rose gradually with increasing dosage of Bromocriptine. Abortion rates were 100% in 0.1 mg and 0.2 mg group. The abortion rate of 0.075 mg group was 75%, with an average embryo number of 2.167±4. 064, which was significantly different from that of control group. As the dosage of Bromocriptine increased, the contents of IFN-γ went up but IL-10, prolactin and progesterone declined in serum and uterus. These results indicate that Bromocriptine has an abortifacient effect on pregnant mice caused by recurrent spontaneous abortion （RSA）. This provides a pragmatic animal model for studying the immune reaction at the maternal-fetal interface [ Acta Zoologica Sinica 52 （3）： 536-541, 2006].
