摘  要:目的观察不同浓度雄激素(睾酮)对体外培养人毛乳头细胞和外根鞘细胞增殖活性的影响。方法采用脱发和非脱发成人头皮标本,用胶原酶消化法分离毛乳头,在角质形成细胞无血清培养基上,对其进行体外培养。采用非脱发成人头皮标本,组织法培养毛囊外根鞘细胞。然后将不同浓度睾酮加入两种毛乳头细胞与外根鞘细胞的共同培养基中,比较其对外根鞘细胞的增殖活性的影响。结果不同浓度睾酮对单独培养的两种毛乳头细胞和外根鞘细胞的增殖性均无作用(与对照组相比P>0.05)。10-9ml/L浓度(与对照组相比P<0.05)、10-8ml/L浓度、10-7ml/L浓度(与对照组相比P<0.01)睾酮与非脱发患者的毛乳头细胞共同培养时,可促进外根鞘细胞增殖。而10-10ml/L浓度(与对照组相比P<0.05)、10-9ml/L浓度、10-8ml/L浓度、10-7ml/L浓度(与对照组相比P<0.01)睾酮与脱发患者的毛乳头细胞共同培养时,对外根鞘细胞出现增殖抑制作用。结论睾酮对外根鞘细胞的影响,与其剂量和毛乳头细胞的存在有关。在这个过程中,毛乳头细胞可能起介导作用。Objective To observe the proliferative activity of human dermal papilla cells and outer root sheath cells in vitro and the results of the interaction with androgen on different concentration. Methods Dermal papillas were isolated from bald and nonbald adults scalp with the method of dissection by collagenase digestion, then the dermal papilla cells were cultured in vitro with keratinocyte cells medium without serum. Out root sheath cells from nonbald adults were cultured in vitro scalp. The testosterone on different concentration was added in the outer root sheath cells which was cocultured with the dermal papilla cells from the bald and nonbald adult scalp and then the proliferative activoty of the outer root sheath cells were examined. Results Testosterone showed no effects on proliferation of either dermal papilla cells or outer root sheath cells cultured alone(P 〉 0. 05 versus control). 10^-9 ml/L concentration of testosterone (P 〈 0.05 versus control). 10^-8 ml/L and 10^-7 ml/L concentration of testosterone( P 〈 0.01 versus control) stimulated proliferation of outer root sheath ceils cocultured with dermal papilla ceils from the nonbald scalps of adult, and inhibitied proliferation of outer root sheath ceils cocultured with dermal papilla ceils from the bald scalps of adult. Conclusion The doage of testosterone and existing of dermal papilla ceils had an impact on the proliferative activity of outer root sheath ceils. Our data indicated that dermal papilla ceils may play a intermediary role in this process.
