摘 要:试验对20个基因型的结球甘蓝材料进行游离小孢子培养。探讨基因型、蔗糖浓度、激素配比与浓度、活性炭等因素对结球甘蓝小孢子胚发生能力的影响。实验结果表明:基因型、蔗糖浓度对胚发生能力影响最大,除个别基因型对蔗糖浓度有较高的要求外,蔗糖浓度在13%、激素组合NAA(0.05mg/mL)+BA(0.1mg/mL)的培养基适合较多基因型的小孢子胚发生。在培齐基中滴加活性碳(10mg/L)每皿1—2滴,诱导效果明显优于不加活性炭。实验中共有11个基因型的甘蓝材料获得了胚状体,并成功筛选出基因型G8小孢子胚高频发生的最佳蔗糖与激素浓度配比。Isolated microspores of 20 genotypes of head cabbage were used for tissue culture to study the effects of genotype, sucrose concentration, combination of hormones and their concentration and activated carbon on embryogeny. The results showed that genotype and sucrose contents in the medium had the greatest influence. For most genotypes in the experiment, a sucrose concentration of 13% and a hormone combination of NAA 0. 05 mg/mL + BA 0. 1 mg/mL gave the optimum result. Addition of 1 or 2 droplets of activated carbon ( 10 mg/mL) to the medium improved embryogeny. Embryoids were induced in 11 cabbage genotypes. The optimum sucrose concentration and hormone combination for the genotype G 8 were identified.
