摘  要:目的观察通脉宁胶囊对血管紧张素Ⅱ(AngⅡ)、溶血磷脂酰胆碱(LPC)诱导的兔血管平滑肌细胞(VSMC)c-fos、c-myc癌基因表达的影响。方法组织贴块法培养兔主动脉平滑肌细胞,用AngⅡ、LPC诱导VSMC增殖,采用RT-PCR方法,观察通脉宁全方及益气拆方和活血拆方药物血清对AngⅡ、LPC诱导的VSMC c-fos、c-myc蛋白表达。结果AngⅡ、LPC组c-fos和c-myc蛋白表达增加,与空白组比较有显著性差异(P<0.01);通脉宁全方组、益气拆方组、活血拆方组均可使c-fos和c-myc的蛋白表达下调,与AngⅡ、LPC组比较有显著性差异(P<0.01);通脉宁全方组与益气拆方组、活血拆方组比较也有显著性差异(P<0.01)。结论通脉宁胶囊可下调AngⅡ、LPC诱导的VSMC c-fos、c-myc基因表达,而对VSMC增殖起到抑制作用。Objective To observe the influence of Tongmaining Capsules on the expressions of cancer genes, c-fos and c-myc, in rabbit vascular smooth muscle cells (VSMC) induced by angiotensin Ⅱ (AngⅡ) and lysophosphatidylcholine (LPC). Method To culture rabbit VSMC by tissue-sticking method and than to induce the proliferation of VSMC by AngⅡ and LPC. To use RT-PCR to observe the influences of the serum of Tongmaining Capsules, its decompositions with the effect of replenishing qi or the effect of activating blood circulation, on the expressions of c-fos and c-myc genes in VSMC. Result The expressions of c-fos and c-myc genes were increased in the AngⅡ group and the LPC group with a significant difference compared with that in the blank group ( P 〈 0.01 ). In the group of Tongmaining Capsules and groups of decompositions with the effect of replenishing qi or the effect of activating blood circulation the expressions of c-fos and c-myc genes were decreased with a significant difference compared with that of the AngⅡ group and LPC group ( P 〈0. 01 ). There was also a significant difference among above three groups(P 〈0. 01 ). Conclusion Tongmaining Capsules has a down-regulative effect on the expressions of c-fos and c-myc genes in VSMC induced by AnglI and LPC, therefore, it can inhibit the proliferation of VSMC.
