摘 要:目的:观察肾疏宁防治肾小球硬化的作用机制。方法:采用单侧肾切除并阿霉素尾静脉注射建立大鼠肾小球硬化模型,观察肾疏宁给药8周后肾小球病理表现,免疫组织化学染色法观察肾小球内α-平滑肌肌动蛋白(α-SMA)、纤维连接蛋白(FN)、层粘连蛋白(LN)、Ⅳ型胶原(ColⅣ)阳性表达,并进行图像分析。结果:肾疏宁治疗组肾小球内α-SMA、FN、LN、ColⅣ和系膜基质较模型组显著减少,与苯那普利无显著性差异。结论:肾疏宁能抑制肾小球系膜细胞表型转化,减少细胞外基质蓄积,对阿霉素肾小球硬化大鼠肾脏具有保护作用。Objective: To explore the mechanism of shenshuning prevent and treat glomerulosclerosis. Method: Glomerulosclerosis model rats were induced by unilateral nephrectomy(Nx) and Doxorubicin injection, randomly divided into 4 groups, Morphological expression changes was observed by lightscope, immunochemical stain to explore the expression of α-smooth muscle actin(α-SMA) and fibronectin(FN), liminin (LN), collagenⅣ (ColⅣ), and analysis with image analysis system. Results: Shenshuning can attenuate α-SMA, FN, LN, ColⅣ expression in renal tissue, decrease messangial matrix. The effective was similar with benazepril, They are significant differentiation with glomerulosclerosis group. Conclusion: Shenshuning can attenuate messangial cell transdifferentiation, decrease messangial matrix deposition, therefore protect glomerulosclerosis renal and prolong pathology development.
