摘 要:目的:观察川芎内酯A预处理对体外培养乳鼠心肌细胞缺氧/复氧(hypoxia/reoxygenation,H/R)损伤的保护作用。方法:单纯低氧/再给氧为心肌细胞低氧2小时、复氧1小时。低氧预处理为心肌细胞低氧25min/复氧30min后,再低氧2小时、复氧1小时。川芎内酯A预处理为心肌细胞经药物孵育55min后,低氧2小时,复氧1小时。实验结束后取上清液测乳酸脱氢酶(LDH)、丙二醛(MDA)、超氧化物歧化酶(SOD);β液体闪烁计数仪测心肌细胞中丝裂原活化蛋白激酶(MAPK)活性。结果:川芎内酯A预处理可使上清液中LDH漏出明显减少,MDA含量降低,SOD活性提高。还可降低心肌细胞内MAPK活性与H/R组比较。结论:川芎内酯A预处理对体外培养的乳鼠心肌细胞缺氧/复氧损伤具有保护作用,MAPK信号传导途径可能参与这一保护作用。Preconditioning protection by chuanxiong-pathalide A was investigated in cultured neonatal rat eardiomyocytes. The results demonstrated that chuanxiong-pathalide A 1 × 10^-7 和 1 × 10^-9mol/L increased the survival rate during reoxygenation 60min following hypoxia 2h. The rise of LDH and MDA significantly decreased and the activity of SOD significantly increased, meanwhile, intervention of MAPK in the precondition- ing was found. In conclusion, preconditioning by chuanxiong-pathalide A demonstrated protection on release of LDH, MDA and SOD in hypoxia / reoxygenation in vitro model. Therefore, it was suggested that chuanxiong-pathalide A may mimic IP by which protecting actions were achieved. The mechanism is likely related with MAPK signal transduction pathway.
