摘  要:目的探讨六味安消胶囊及其组分对豚鼠结肠黑变及动力学影响。方法 20只豚鼠随机分为4组:正常组、大黄组、诃子组和六味安消胶囊组,后三组分别给予不同药物灌服30 d。处死动物,检测炭末推进率,观察结肠黑变情况,并对盲肠及近段结肠末染色切片行图像分析。结果大黄组豚鼠结肠有黑变,以盲肠及近段结肠为著。病理改变类似人类。大黄组(87.71%±25.87%)、诃子组(67.63%±19.05%)及六味安消胶囊组(91.14%±16.13%)炭末推进率较正常组(40.38%±6.99%)明显增快(P<0.05)。图像分析显示,诃子组、六味安消胶囊组盲肠、近段结肠固有层色素颗粒灰度及阳性面积比与正常组差异无统计学意义(P>0.05)。大黄组盲肠、近段结肠固有层色素颗粒灰度(盲肠133.18±1.72,结肠137.40±1.39)及阳性面积比(盲肠4.65%±0.76%,结肠2.36%±0.42%)与正常组差异有统计学意义(P<0.01)。结论大黄可致豚鼠实验性结肠黑变病,六味安消胶囊致结肠黑变作用不明显,诃子有预防结肠黑变作用。Objective To study the effect of Liuwei Anxiao capsules and its ingredient on nigrities and motility of colon of guinea pig. Methods Twenty guinea pigs were randomly divided into 4 groups: normal group, rhubarb group, fructus group and Liuwei Anxiao capsules group. Except the normal group, the other three groups received a daily intragastric administration of different drugs for 30 days. The animals were then killed, intestinal propulsive rate of carbon and nigrities of colon were observed. Image analysis was performed in unstained sections of cecum and proximal colon. Results Macroscopically, the mucosa of the cecum and proximal colon was brown in color in the animals in rhubarb group. Pathological change closely resembled human melanosis coll. Intestinal propulsive rates of carbon were significantly faster in rhubarb group (87.71±25.87%), fructus group (67.63± 19.05%) and Liuwei Anxiao capsules group (91. 14 %±16.13 %) than that in normal group (40.38± 6.99 % ) (P 〈 0.05). Image analysis showed that the grey scale and area % of pigments of cecum and proximal colon in fructus group and Liuwei Anxiao capsules group was similar with that of normal group. The grey scale(cecum 133.18±1.72, colon 137.40 ± 1.39, P 〈 0. 01) and area % (cecum 4.65 ±0. 76%, colon 2.36%. 42 M, P 〈0. 01) of pigments of cecum and proximal colon in rhubarb group was different with that of normal group. Conclusions Rhubarb can induce melanosis coli in guinea pig. The effect of Liuwei Anxiao capsules on nigrities of colon is not significant. Fructus can prevent colon from melanosis coli.
