摘  要:目的:分析硝酸酯类药物预适应对不同部位的心肌细胞缺氧再复氧的保护作用。方法:实验于2003-05/2005-09在泰山医学院中心实验室完成。取新生的Wistar大鼠100只,无菌取出心脏,培养不同部位的心肌细胞(心房、左心室、右心室),分为4组:①空白对照组:不作任何处理。②单纯缺氧/再复氧组:在含有体积分数为0.95的N2、0.03的CO2和0.02的O2混合气体的缺氧培养箱内孵育2h,正常培养箱内孵育1h。③L-精氨酸处理+缺氧/再复氧组:先用10mmol/LL-精氨酸处理细胞2h,然后进行缺氧再复氧。④缺氧预适应组:先将细胞缺氧15min,再复氧15min,循环4次,然后进行缺氧再复氧。观察各组细胞坏死率、细胞凋亡情况、培养液中乳酸脱氢酶浓度及细胞内游离钙离子浓度等。结果:①单纯缺氧/再复氧组不同部位的细胞坏死率、培养液中乳酸脱氢酶释放量及胞内钙离子浓度均显著高于空白对照组(P<0.01),不同部位间比较差异无显著性意义(P>0.05)。②缺氧预适应组、L-精氨酸处理+缺氧/再复氧组不同部位的细胞坏死率、培养液中乳酸脱氢酶释放量及胞内钙离子浓度均显著低于单纯缺氧/再复氧组(P<0.05)。结论:L-精氨酸的预适应同经典的缺氧预适应一样,可以对缺氧再复氧损伤产生明显的保护作用,其可能是通过降低心肌细胞内钙离子的浓度来发挥作用的,而不同部位的心肌细胞产生药物预适应的能力无明显差别。AIM: To analyze the protective effect of nitrate medicine preconditioning on the hypoxia/re-oxygen of different parts of the myocardial cells. METHODS: The experiment was performed at the central laboratory, Taishan Medical College between May 2003 and September 2005. Totally 100 neonatal Wistar rats were selected. The hearts were isolated under sterility condition. Myocardial cells (cardiac atrium, left ventricle and right ventricle) from different parts were cultured and assigned randomly into 4 groups: (1)Blank control group: The rats were not treated with any disposal: (2) Simple hypoxia/re-oxygen group: The rats were incubated in the anerobic incubator contained mixed gas containing N2 of 0.95 volume fraction, CO2 of 0.03 volume fraction and O2 of 0.02 volume fraction for 2 hours, and then incubated in normal incubator for 1 hour. (3)L-arginine plus hypoxia/re-oxygen group: The cells were disposed for 2 hours with 10 mmol/L L-arginine, and then treated with hypoxia/re-oxygen. (4) Hypoxia preconditioning group: Cells were treated under hypoxia for 15 minutes, and then re-oxygen for 15 minutes and circulation for 4 times, followed by hypoxia/re-oxygen. Cellular necrosis rate, apoptosis, concentration of lactic acid dehydrogenase in culture solution and concentration of free calcium ion and so on were observed. RESULTS: (1) Cellular necrosis rate, concentration of lactic acid dehydrogenase in culture solution and concentration of free calcium ion of different parts in the simple hypoxia/re-oxygen group were significantly higher than those of the blank control group (P 〈 0.01 ). There was insignificant difference in different parts (P 〉 0.05). (2)Cellular necrosis rate, concentration of lactic acid dehydrogenase in culture solution and concentration of free calcium ion of different parts in the hypoxia preconditioning group and L-arginine plus hypoxia/re-oxygen group were significantly lower than those of the simple hypoxia/re-oxygen group (P 〈0.0
