摘  要:目的观察葡萄糖神经酰胺合成酶(glucosylceramide synthase,GCS)基因在人白血病细胞的表达及其与肿瘤多药耐药的关系。方法采用RT-PCR方法分析耐药/非耐药白血病患者GCS基因和多药耐药(multidrug re-sistance,MDR)基因(mdr1)的表达水平及其两者的差异。结果白血病耐药组GCS基因扩增条带光密度相对比值明显高于非耐药组(P<0.05);白血病患者标本GCS基因扩增条带显著增强时往往伴有mdr1基因的表达,两者呈正相关(P<0.01、γ=0.6)。结论白血病多药耐药患者GCS基因的表达明显强于非耐药患者,GCS基因的表达与mdr1基因相关。提示GCS基因可能在白血病多药耐药中起着重要作用。Objective To investigate the difference of glucosylceramide synthase (GCS) mRNA expression between drug-sensitive and drug-resistant human leukemia and to study the relationship between glucosylceramide synthase (GCS) mRNA and multidrug resistance gene (mdr1) in drug-resistant human leukemia. Methods Reverse transcriptional polymerase chain reaction (RT-PCR) was employed to analyze the expression of GCS and mdr1 mRNA in drug-sensitive and drug-resistant human leukemia. Results The comparative optical density ratio of the amplified straps in the drug- resistant group were significantly higher than one in the drug-sensitive group ( P 〈 0.05). There was positive correlation between the expression of GCS gene and mdr1 gene in leukemia samples (P 〈 0.01, y = 0.6). Conclusion GCS gene may play an important role in MDR during tumor progression.
