摘 要:目的探讨脑损伤后c-fos基因和蛋白的表达变化规律与脑水肿之间的关系。方法利用Feeney's自由落体法制作局灶性脑损伤动物模型,应用原位杂交和免疫组化技术检测不同受伤时间段c-fos mRNA和蛋白的表达,同时观察脑组织的水肿情况。结果c-fos mRNA在伤后15 m in即有高水平表达,并出现两次表达高峰,分别出现于30 m in和16 h。c-fos蛋白随受伤时间的延长表达逐渐增强,在16 h表达最强,随后表达下降,48 h时表达又有升高现象。脑水肿在伤后30 m in开始发生,逐渐加重,6 h达高峰,持续至16 h,后逐渐减轻,48 h基本正常。结论脑损伤后c-fos的首次表达高峰可能参与了损伤神经元的修复和生存,与神经细胞的保护机制有关。c-fos第二次表达高峰与脑水肿所致缺血缺氧有关,参与脑损伤后的继发性损害。Objective To explore the association of c-fos expression and brain edema in the experimental brain injury. Methods Rat focal brain injured models were made using Freeney's method. The expressions of c-fos mRNA and protein were detected by situ hybridization and immunohistochemical method in various periods after brain injury. Results The expressions of c-fos mRNA were identified as early as 15 minutes, and reached maximum levels in 30 minutes and 16 hours after injury. The protein expressions of c-fos were up-regulated according to injury time, and reached maximum in 16 hours, and decreased lately, however, increased in 48 hours. Brain edema occur in 30 minutes after injury,while water content got its peak after 6 hours to 16 hours. Conclusion The first expression maximum of c-fos may have neuroprotective effect on the traumatic rat brain. However, the second expression maximum has correlation to ischemia induced by brain edema, and participates in the secondly brain injury.
