摘  要:目的研究胡麻油烟冷凝物致人胚胎二倍体细胞恶性转化作用.方法以人胚肺二倍体细胞株为靶细胞,建立体外转化系统,以胡麻油烟冷凝物作为受试物染毒人胚肺二倍体细胞,观察细胞转化情况,并对转化细胞进行生物学性状检测和鉴定.结果实验剂量范围内的胡麻油烟冷凝物(25~400μg/mL)能够诱导人胚肺二倍体细胞发生失去接触抑制性、饱和密度增大、血清需求降低,ConA凝集能力增强、锚着依赖性丧失等多种与肿瘤形成相关的表型改变,并且剂量-反应关系明显(r=0.972,P<0.01).结论胡麻油烟冷凝物具有诱导人胚肺二倍体细胞转化的作用.Objective To study the potential carcinogenic risk of siritch oil fume (SOFC) for human embryo lung diploid cells. Methods Human embryo lung diploid cells were subjected to establish transformation system in vitro. Different concentrations of SOFC were added into the DMEM cultures and co-incubated with human embryo lung diploid cells. The malignant degree of transformation was observed by assessing the biological characteristics of the cells. Results The concentrations of SOFC (25 - 400 μg/mL) could induce the malignant transformation of the human embryo lung diploid cell, such as loss of contact-dependent inhibition, saturated density augmenting, growth at low serum concentration, agglutination by different concentration of ConA, loss of anchorage dependence, etc. Those characteristics associated with malignant transformation and with an obvious dose-response relationship. Conclusion SOFC could induce human embryo lung diploid cell malignant transformation.
